Mexicana A mycelium
    #3275936 - 10/25/04 06:00 PM (12 years, 4 months ago)

I just want to know if this looks OK:

because this does not look like (i think) any of the mexicana mycelium photos or descriptions of it that I read (not only white but shady or something..)
It looked ,compared to pan cyan and ps. cubensis mycelium on agar,
very dense and thick and very white. and there is no sectoring visible at all, it was a multispore syringe.

can anyone who has done this before say "that is mexicana A"?
or is that a stupid question?

thanks

I think it looks good. It will form sclerotia and change the color the next time.

bought it from azarius.com

it is the only dish out of 10 that started some growth. I am
planning to make more dishes out of it and then do some transfers
to jars with rice and autoclavable bags with rice. I cant get the
water amount of grass seeds right and I've read rice is also very
good.

In my culture it forms very fast sclerotia in the petri dish. Few days then it begins.

Only one plate out of 10 grew anything? That suggests very few viable spores in the syringe and could explain the lack of sectoring. Hopefully its not a monospore culture. Is there a slimy pink contaminate in that plate or is it just the lighting?

thats the light I guess..there is no contam as far as I see.
I hope its not mono too..but no way to check it out I guess.
I will have to wait and see right?
It was a sporeprint and I made a syringe of it. Waited for ten days
before using it...and yes only one plate grew something

anymore comments from anyone?

Please say something [Re: nimbus] * 1
    #3300290 - 10/31/04 07:02 AM (12 years, 4 months ago)

Is there no way to tell wether this is monokaryotic or dikaryotic, other than having to
wait and see if it will fruit? Becuse I am planning to divide some of it on new dishes
and then go on trying to produce sclerotia..

My biggest concern is, i repeat: it was a multispore syringe..but no sectoring at all?!!

Here's how it looks today:

I think if you look at that growth under a microscope, you'll see a ton of bacterial rods attached. Try pouring hot antibiotic agar right on top of the growth. Fill up the dish. I've been having good luck cleaning up cultures lately that way. The hot antibiotic agar seems to kick the contams in the ass, but the myc survives and grows up through the layer to the top. When this happens, use your scalpel and gently 'shave' some myc off the top without getting any of the agar. Transfer a very small piece of mycelium to a new dish, and when it grows out a bit repeat the process. It takes a time or three to get a good, clean culture. Try it.
