7 out of 22! WTF!
    #3246663 - 10/12/04 11:10 AM (12 years, 4 months ago)

OK, here's the deal. About a month ago, I made up 22 half-pint jars and PCed them at 15psi for 60 minutes. Two weeks went by, no contams, so I inocced. The jars are 90-95% colonized. However, 15 of them show small amounts of green in the uncolonized parts. That sucks, I sterilized the room and sterilized the needle between jars. My syringes came from a reputable shroomery sponsor. Should I wait to see if the mycelium takes over or just keep the 7 that show no signs of contamination? What could possibly be going wrong? Could waiting too long after PCing to inoculate be my problem? Thanks.

Why did you INOCULAT 2 weeks later?

You should INOCULAT when the jars have cooled.

I read on fanaticus.com that you could test the jars by letting them sit before inoculation to see if you are PCing correctly. I'm not going to do that again. Do yo think that could've been my problem?

If the contamination starts directly at the inoculation points and shortly after inoculation, then the syringes(either dirty needle or diry content) are to blame. Otherwise not.

heres what COULD have happened.

when u let the jars sit for the 2 weeks.. were the holes covered? contams may have entered through there.

also what kind of temp etc. conditions were the jars in for the 2 weeks? was it a controlled enviroment? or were they jus sittin in a box or what? those could have also been factors in it.

I had tape covering the holes. The jars were in a box probably at about room temp.

I think on my next run, I'll omit the observation stage and inoculate right after cooling. I'm also making a tub in tub incubator that should be ready. Hopefully the myc will outgrow any contams.
So tell me, should I throw out the jars that show a small amount of green or see if the myc out competes the contams? They are about 90-95% colonized.

I'd say yeah. Don't mess with contam. It's probably worse inside anyways. If you decide to disect it, do it outside. Don't mess your pad up with contam. Good luck.

Letting the jars sit isn't causing the contams. You should be able to let em sit for a year. They can't contam if there isn't some kind of contam spore in there.
