Successful mycelium transplant

on aug 03 i started 12 amazoinan jars, as of the end of Sep only 2 colonized

i decided that i might as well ag do a little experementing. i sterilized my suurounding and tools and went to work. i birthed the two colonized cakes along with one completly un colonized cake. using a razorblade i cut a 1 inch cone out of the bottom of the uncolonized cake. i then removed the whispy mycelium from the sealing vermicluite layer from the colonized. i then placed the whispy mycelium inside the depression created in the un colonized cake and placed it back into the jar and resealed it. roughly 10 days later the transplant jar is 1/2 colonized with absouletly no contamination. a successful transplant. :thumbup:

Thats a transfer. Ive done G2G many times with grains. With good cube growth you should have colonized jars in 5-8 days.

    Quick question on G2G's.  I did one set from two rye pints, distributed them to several other pints.  Worked fine.
    Then, I tried to do a transfer from some quarts to other quarts.  This failed and I got wet spot in the grains.  I *think* that the problem was either a) the colonized myc/substrate that was transferred was going on 4th generation; or b) for some reason the bacillus endospores survived the PC (I did soak 36 hours to allow germination prior to PCing).
      My question is: should you split up the chunks of colonized substrate when G2Ging to make more colonization points in the new jar?  I just had an essentially fully conlonized rye jar that I shook and the myc never really recovered.  :mad:  So now I'm hesitant to break up the nice jar of thick white colonized substrate when I G2G it.  Should I just toss a chunk in the new jar and that should do it? 
      Okay.  Sorry this post was a little convoluted.  I just woke up and my mind's not out of dreamland yet.  THanks! :grin:

bad news, the transplant cake was overwelmed by black contamination overnight last night, it is completely lost, on the other hand the spore print i made allowed me to inoc many more jars so this wasnt a total lossi beleive the whole problem was the low count of spores in the original syringe

That was not the problem. There were plenty of spores. I don't know if it takes one or two (I'm pretty sure two) but no more then that are needed.
