tape on or off?

sorry for my recent newb questions but i couldnt find in the PF tek if u leave the tape over the holes after innoculation or keep it on. I cant tell if the substrate is moist enough and am afraid taking the tape off would let neeeded moisture out? should i wait until after i see some white fuzz to take the tape off or take it off now ???

I leave the tape on until the cake is about %80, then i take it off and flip the jar over for invitro..

that is how I do it and never have had any problems with contamination....

Fuck that, don't use tape at all.

Keep foil over the top and leave it at that.

I dont imagine you would lose a lot of moisture from just the jar holes. I believe it is better to have the tape off. The top dry layer of verm wards off airborn contams, and there should be enough water in your mix to survive. However, if it makes you nervous and you're worried about it, seal those puppies up and check on em in a few days. Always make sure you are happy with your substrate before wasting syringe solution on substrate that is too dry, or too wet.
When colonization begins and spreads, you will need to loosen the lid for gas exchange.

i dont use tape either. and i am having great growth and no contam (knock on wood)

You don't have to remove it at all, but if it's 80% colonized or more there is a pretty good chance you'll be alright. If you want to stay completely safe you might want to wait slightly longer though...

Couldn't agree more! Think of it as an improvment to the PFTek hell with the tape foil holds moisture in, makes for a nice contam barrier and you can write on it!!!
