pressure cooking

we pressure cooked our pf jars for 20 mins. my question is we put the lids on upside down and screwed the lid down lightly so there was a very tiny bit of pressure. should we have screwed it on so there was no pressure at all. or is it ok the way we did it?

also we only have 70% isopropyl alc. do we need 99% to be sterile when injecting?

I was under the impression the pressure cooking process should last about an hour, but im new so dont take my word for it. If there were holes in the lids of the jars, there is practically no way that the pressure would not build up inside them, but i think leaving them lightly screwed on as you did is the proper way. AS for the alcohol, i would imagine 70% would work, but again, im new so take it for what you will...

you should pc for 1 hour. leave the lids 1/4 to 1/2 a turn open.

im doing pf tek. and it said 15 mins of pressure cooking. we microwaved the substrate for 5 mins before putting it in the jars and we were extremely sterile. we turned our kitchen into a clean room

u think we should do them again for an hour but pc for and hour this time?????
we could only do 3 jars at a time and we have 12 jars that is why we decided on 20 mins.


i dont want contams. is it possible to let the jars sit for 5 days to see if contams show up before we inoculate?

I agree 20-30min. at 15lbs. is very safe for PF jars. Also snug down lids while PCing then when cooled remove the foil and tape then innoculate leave the tape off (if you even used it in the first place) crack open the lid 1/4-1/2 turn as stated above (really aids in gas exchange :wink: ) place the foil back on and incubate at 82-84F. and you'll be on your way!!!
