colonize without pressure cooking?

Is the only reason for pressure cooking to Sterilize? Or does the rice flour need to be cooked. I know that it will probably get contaminated without cooking, but is it possible to actually colonize without pressure cooking?

It's a little of both. PCing sterilizes the substrate, but it also softens and moistens your grain. If you start with sterile grain and soak it (to soften) in sterile conditions, the mycelium will colonize it, maybe more quickly than if you PC, though I've never tried this.
