Coffee-soaked grain; tan myc?
    #3137132 - 09/15/04 08:40 AM (18 years, 2 months ago)

So, I'm pretty sure I'll toss these jars and start again.  What I'm wondering is if this is not normal or not.  I used a karo culture to innoc some jars.  Alas, I didn't wait long enough for the myc. to colonize the liquid.  So, only a small amount of myc was injected into the jars.  So, the myc is colonizing the jars very slowly.  I actually turned off the heat in the incubator a couple of days ago, thinking I would just be breeding contams.  But, now, after checking, I'm not sure.  Is off-color myc (tan/beige) normal for coffee-soaked grain substrates?
I just don't want to waste another few jars with this Karo culture.  It's been about 2 weeks since I innoculated these jars.  I see nothing especially nasty, it just looks like the myc was too sparse to colonize well.  Now, it actually looks as though whatever is growing is finding its sea legs.  Any ideas on this one??  Thanks very much!

Sort of looks like the pics of Wet Spot (bacillus) bacteria, first on this list:
the contams list

That or it's the coffee color coming out. But probably the former. Does it smell?

thats definately bacterial from an excess of water...

Gnubobo, next time, add about 1gram of finely ground gypsum
and shake it well before you sterilize, that should help.

Thanks all. Yeah, there were a bunch of little white dots in the grain, then those sort of disappeared and now there are parts that are sort of oozy. The more I think about it, the more I think these should be discarded posthaste.
By the way, Prisoner#1, why is that extra moisture allows bacteria? Can bacteria get through tyvek and then grow in a wet environment? I'm guessing because the myc never colonized the grain was prone to bacterial growth? Is this correct? Thanks.

Yeah, I checked out the Bacillus link. That pretty much sounds/looks like what I've got here.
Is there any reason why a coffee-soak would prevent bacterial endospores from germinating? I did soak these for 24 hours before PCing. I didn't have a problem when I did the same thing last time with rye. Hmmmm.
Thanks!

When RR does rye (the guy who was doing this coffee tek thing big time) with coffee he PC's for 2 hours or so IIRC. Coffee makes EVERYTHING grow quicker, including contams. So you have to PC longer to kill more of them bad suckers. They look like they could be a little dryer too...
