Cloning in liquid culture, is it possible?
    #3131003 - 09/13/04 06:36 PM (20 years, 4 months ago)

hello. Ive recently been reading up on cloning with cardboard and also making liquid cultures. Can you put a piece of mush flesh directly in a sterile karo solution to clone it? or should you clone with cardboard and use a piece of that? Seems like you could do it.

I have done this with just sterile water no need for Karo unless your looking at some kinda long time storage. I just take a very nice chunk (1-2") as long as possible and dropped it into my homemade Eberbach which was PCed with h2o in it. It makes for some nice quick cloning. I also use CBS's (core biopsy syringes) after Mycota (6T) turned me on to them a few years back. They make taking sterile samples a breeze!!!!!

That may be fine for people with a fancy blender, but you can also just drop a small piece in some 3% malt extract solution and let it grow up.

CBS's are used for taking sterile tissue samples from organs, tumors, mushrooms, etc; They are also reusable.

When available I've seen heard them go from $35 on up to $75 each. I do have a source but it's inconsistant so basically I put my order in and wait, last time it was almost 3mos. When I get them they go for $35+shipping (2-3 day delivery). These shoot the needle out and back into a sheath keeping the sample from ever being exposed to an open environment, I like to store the syringe in a bottle of alcohol right up until the sample is taken.

Could we see the homemade Eberbach? Possibly on a new thread with pointers and info? It could even be added to the tek's

Just looked and it must be in storage, when I get a chance I'll grab it and take a pic for ya guys  It's really quite simple, it is a regular mouth pint jar screwed to a blender base (poly carbonate), not all bases work, my first deformed badly (Proctor Silex) but the second one (Oster) worked flawlessly. The base was drilled and tapped with a small machine screw which is where the syringe is inserted after the screw is removed. It works great when used with the CBS (core biopsy syringe!) take care guys
