Karo jar cloudy yet full of myc bits.
    #3133037 - 09/14/04 03:58 AM (12 years, 2 months ago)

So, this karo jar is cloudy.  I assume there is contamination.  But the other two karo jars have been showing bits of myc, these bits settle,; when the jar is swirled, we see bits of myc all over.
Now, the first jar is showing more bits of myc (thought it was contamed?, as it was cloudy).  Beyond this, the other jars with bits of myc floating around are looking more cloudy.
Hmmm....these jars are tyveked and sealed.  I don't understand this development.  Is this normal?
I swirl the water/myc/karo in the jar and things get cloudy, tho it is quite obvious there are mycellium bits floating around like winter wonderland.
Any help??  Many thanks in advance!

Some of the corn sugar in the syrup usually carmelizes/crystalizes during sterilization and makes a heavy perhaps golden colored sediment (but it can appear white, if the crystals are small). It sounds like you just have some of that, since it settles out right away and goes back to mostly clear. Most contams will either float very similar to how mycelium does in the case of molds, or will be mostly invisible in the case of bacteria (unless it turns some of mycelium black, in which case it's floating exactly like the mycelium except it's black).

If you can see the sediment in its settled form at the bottom it should look sort of yellowish/golden since the light is then shining through several 'layers' of the material.

If it is indeed this, which is probable, then you're fine, especially if growth continues. How much karo vs water did you use?

A sure way to tell would be to plate out some of the solution on agar.

Otherwise, from the description I would say that the first jar is definately contamed, but the other jars are probably fine. If the solution remains cloudy and doesn't settle out then it is surely contamed, but if it settles out it's likely just poor media to blame.

It is not unknown for vendors(PF did) to send out syringes that are contaminated with yeast. It only happened once as far as I know, but who knows how many got sent out that way.

Next time try a better media like PD or malt.

Why what?

He had a yeast contamination problem and sent out a ton of syringes with yeast contamination. It happens.

cloudy LC is a certain sign of contamination.

most likely bacterial.

either way, your mycelium is toast and so is that jar.

start again.

I noticed some of my jars got slightly cloudy. A couple got really cloudy and I tossed them. But the slightly cloudy ones I injected with some h202. Now the jars look more clear. But on some of them there is a settled white dust that clouds the water when disturbed. Could this be dead bacteria? There is plenty of myc in the jars, but I'm not sure if it's still growing. Is it possible to kill the bacteria without killing the myc?

from my experience i have had lc that looked great and were contam and ones that look like crap and did every jar i could find.
i usually do a test jar or two inoc those jars wait 3-4days if they are contamed u will know and u could open one and smell it for bacteria. lc has burned me in the past pc 24 jars inect and bam all bad so now i test every lc to save me all the pc time.
