Karo Question

I tried ijecting a few jars with some karo i made. There was chunks of white stuff floating around and i even sucked some of it up. Problem was i got no growth whatsoever.

So, say you inject a jar of karo with spores and wait two weeks. there is fluffies floating, but only little bits. Now, do you need to suck up those bits to be useable, or will any of the water work?


Also, why, in the karo jars i shot mycelium into turned cloudy, and the ones i shot spores into did not, but still have significant white fluff floating around. Normal?

#1 those "clouds" need to be broken up then drawn up. A couple dime sized clouds broken up good is all ya need for some hot liquid innoc.!!! GL

Cloudiness is likely a sign of yeast or bactreial contamination. Throw em' out.

Hyphae answered that one well, you want to break up the mycelium and inject that. If its cloudy or opaque then it may be bacterial or yeast as fastfred suggests. Give it a gentle shake and it should look something like this....
your media might be darker or lighter.

Ive let that one culture go a bit too far, and now its going to be a bugger to break up (I cant get a good shot of it now, almost a solid block of 'fluff'), but Ive got access to a laminar flow hood and a big arsed horse syringe. So I'll break it up and make a number of syringes.

ahhhhh, making much more sense now

I was wondering why i had 11/12 contams after a karo injection, yet the jar i used was cloudy. I was origionally told thats what i want.

So, last night i used a jar that was clear with a huge glob of mycelium. And yes, i know now it needs to be broken up, it wouldnt even suck up into the syringe until it was.

Well i feel better because only one of my karo jars went sour so it seems, 5/6 aint too bad. Also, i injected the five good jars with spores, not mycelium water drawn from a cultured jar like the last one. Much better contam rate.

Edit: also, for reference, i hear a lot of people wondering if carmalized karo still works. Mine has gone slightly golden after PC'ing , its growing just fine

Carmalized Karo  depending on how bad retards growth anywhere from a little (off color) to totally inhibits growth (dark amber). The clouds can be broken up as Magash has also suggested by drawing and purging of the syringe repeatedly :wink: Often times after the myc is shredded into a bizillion microscopic pieces the jar will have a cloudy look to it IME. But again we're talking after breakup FF :wink:

You don't seem to understand the difference between "clouds" and "cloudiness". Yeast is one of the most common contaminants and causes the media to turn cloudy.

What's your big fixation with "Faggotry" anyway?

and i knew exactly what everyone in this thread was talking about, except you.
so until you have something constructive (and correct) to tell some one

I'm not flaming you, you really have no clue. If your contam is a unicellular organism like yeast or bacteria you won't be able to see it in your media. It will cause the media to become "cloudy" though... The level cloudiness is called the OD and is a standard measure of bacterial concentration.

Try learning about things before you start spouting off... It makes you look really ignorant.

Here is some "book learnin'" for you...

"Because it is the simplest and fastest method, measurement of the number of bacteria in a culture will be done using absorbance. which can be calculated from transmittance using a Bausch and Lomb spectrophotometer (Spec 20). The principle is that as the number of bacteria in a broth culture increases, the turbidity (cloudiness) of the solution increases. As turbidity increases, less light can be transmitted through the solution as more of the light is absorbed by the bacteria."

god damn, why does this shit happen on seemingly every thread? i swear this place is FILLED with little fuckin babies who like to feel better by being an asshole. It would be really interesting if this were a LIVE face-to-face forum, i bet people would be a little nicer because this much cockyness for such a SIMPLE question gets peoples asses kicked everyday.
