needle cooldown

i searched for information on this but found nothing.
its been 4 days since my friend innoculated 12 1/2 pint jars and there is no signs of growth so far. temps have ranged from 79-86.
when he sterilized the first few jars he flamed the needle and waited probly 20 seconds before injecting. he flamed between each jar for the first 3. he flamed once for the next 4 because he got tired of flaming in between each one. for the next 5 he flamed a couple times and whiped off with a paper towel that was soaked with lysol.
when he was flaming he could hear crackling as the liquid in the needle got hot he thinks.
is it possible that he will have some dead jars because of killing the spores from heat? the needles do disipate heat very quickly though...
how long does everybody wait after a flame if they dont use anything to whipe it off with?

IF it's been 4 days and you see no sign of anything. No worries that means it's sterile, so keep proper techniques to keep it that way. Also 79-86 is a significantly wide spectrum of temps. IT needs to stay a steady 85-86 for proper cultivation. Incubators are so easy to make yourself man. So proper temps is the key to faster colonization.

deffinately wipe w/iso after you flame it. that's what i do and it works great.

Spores will germinate in a wide range of temps warmer the better 82-84 is the optimal range for incubation/germination but you should be just fine Spores take awhile to germinate, as much as 21 days is not uncommon relax in this hobby patience IS a virtue! BTW I wait maybe 10sec. squirt a drop out then innoc. away never reflaming unless the needle hits something questionable. GL

well i have my inncubator set up. temp has been varying because i just got a digital timer for my heat source. ive been manually turning it on and off that you know how that will make it vary. now im playing with the times on the timer to find right ammount of time to keep it at 86

get a "tub-n-tub" with a cheap ass fish tank heater.(50w)
It costs next to nothing to run, and it's constant.

FYI temps inside the jar will be 2 degrees higher bc of metabolic activity still 86 you be just fine.

86 is a little high.... after that growth tends to slow.

im using a 6 foot rope light for a heat source. it is wrapped around a wooden tray with a bevelled edge that my jars sit on(so they dont touch the lights). im growing invitro and it is recommended to expose to light from the beginning. everything fits neatly under my bed in a 6 inch height rubbermaid. i ran it last night 20 min on 80 min off. did ok...turned up to 30 min on 70 min off today. hopefully that will keep the temp range up. the bin is well insulted and it keeps warm for awhile.

thats not enough heat to burn that wood. and if anything did burn its in a sealed container, it would smother itself.

Still scary. If the cord decided to short out (being wrapped against itself raises this possibility) you would get an instant fire and probably it would melt through the tub and get its own air so there goes your 'sealed container can't flame up' theory. But alas you're going to do what you're going to do, and it'll probably be fine but damn I'd be nervous if I were you.
So what strain is it your friend is using? I have some B+ from spores101 and absolutely nothing has happened in the 6 jars I set up with it. However the other two strains I got at the same time and set up at the same time are working very well. I have heard B+ takes time sometimes, but it's been weeks and I have nothing in any of the things I injected with B+, following same procedures as with the others which again are working great.

he got golden teachers from spores101. the free one he got was orissa india. nothing is happening with the golden teachers so far...it was a very very clear syringe. the OI he can see spores in them. he has a feeling they arnt gonna grow... and these rope lights wont cause a fire...when they get their hottest he can hold them in his hands...they arnt that hot. they are coated in a thick plastic. he wonders how good OI will do invitro? edit: found growth in 3 of the jars! my friend is very excited!. also one jar appears to be contaminated. a big brown fuzz has consumed 33percent of the jar.

i think that the orissa is too big of a strain for invitro.....but that is just my opinion.

Cool, yeah my B+ from them was very clear as well, but not the other two which were Blue Meanie (assuming cubensis, not the 'real deal' copes) and Oaxacan. I really like the Oaxacan but need to try some of the Thai/Cambodian ones that grow fast crazy style. Maybe they were/are having syringe loading problems and some come out weak.
True on the ropelights, I didn't realize they were the good style ones (like they use on floors in theaters, and in display cases, and stuff). Those are pretty safe then compared to what I was picturing, the supercheapie lame ones.
I don't know anything about invitro readiness, nor anything at all about OI's, so I leave that up to someone else.

yeah these are good ones. 6 feet of it is a good heat source for invitro. still playing with the timer to get the temps right. i think he still might try out the OI's invitro...if nobodys done it...somebody has to try. my friend is very excited that he got growth signs within 5 days. shit just like exploded overnight.
