chemical sterilization by ethylene-oxide ?
    #2979021 - 08/08/04 06:15 PM (19 years, 7 months ago)

On the package of a sterile plastic syringe there is following written:

Single-use syringe.
Sterilized by ethylene-oxide.
STERILE, non-pyrogenic.
Not to be used with Paraldehyde.
Do not use if package is damaged.

Does anybody know how this sterilizing method works and if it
would be possible to sterilize used plastic petri dishes with it ?

Ethylene oxide is used to sterilize heat sensative media tools like syringes and plastic petri dishes. It works by covalently attaching itself to cell proteins, which makes it a very effective microbicidal and sporicidal. The drawbacks are that it is very dangerous to work with in respect to serious health risks. It is a known carcinogen causing leukimia for one, and it causes a variety of other respritory irritations and complications. Basicaly not something you would want to mess around with unless you have the proper ppe like a positive pressure resperator and fume hood.

It's really a pity that all teks that seem to be promising at the first glance have some drawbacks :frown: .

It's a very bad fire hazard as well. It's used in several FAE-type weapons, where it is preferred over other fuels because even if it fails to ignite, it kills people anyway.
Stick with a PC .

If you have a good glovebox then here is a method that gives pretty good results, but it is a bit of work.

1. Clean the petri's well with soap and water to remove any residual media. Make sure the water is very hot to dissove any agar left over.

2. Place the dishes into the glovebox after you have thoroughly cleaned and dissenfected the inside. Place a bowl of 20% bleach/water solution and a bowl of 70% isopropyl alcohol inside the glovebox as well.

3. Place as many of the petri halves into the bleach solution as can fit and still be submerged and let them sit for about five minutes.

4. Remove them one at a time and dunk them in the isopropyl for a good few seconds to remove any residue from the bleach and place them aside to dry.

Repeat the process until they are all washed and as long as the glovebox is sterile you wont re-contaminate the dishes. Placing a few new ziplock baggies in with the other items at the beggining will give you a good way to store them once they are dry. Just put them in the ziplocks before you open the glovebox and they will stay contam free until you need them.

Good Luck!

Forget about using ETO at home.....

I found this snippet at http://www.reproline.jhu.edu/english/4morerh/4ip/IP_manual/11_Sterilization.pdf

Ok, I am convinced that it is not the best method for a home grower .
Think I will try out Mykey's method.

FAE = Fuel-Air Explosive

Like a mix between a bomb and a flamethrower. Bomb shell pops above target scattering mist of burning fuel over target and certain range around target as well. Think napalm, but more up-to-date and more volatile.
