I have used horse manure from commercial farms with no problems. Likely they are given antibiotics, but not specifically antifungals. Most of these things are pretty well metabolized within the animal, the amounts used are small in comparison to the amounts of feed and water the animal ingests. The manure should be "leached" before use, meaning it is rinsed, and cooked in the sun to remove ammonia. It is essentially composted at this point. The sun will break quite a bit of stuff down. It should be dry and not smell like shit, and should break apart easily. I've put manure from the farm that was a mix of fresher and older in planting trays with drain holes at the edge of my garden, watered them lightly and let the sun cook them for a good amount of time till composted and dry, and never had issues with using it. That being said, its questionable if it is necessary or useful. I ran a culture of PE in side by side tubs, one with coir, one supplemented with manure. The manure tub yielded slightly less and HPLC tested slightly lower. Keep in mind this is a sample size of n=1 and my opinion is that if you ran this experiment 1000 times, the results would probably even out to negligible. I have also used alpaca, cow, deer and elk manure, and haven't noticed any amazing or horrible results from any of these experiments. Solid culture work, good technique, pasteurizing simple coir substrates, and of course, throwing out anything that doesn't look 100% at every stage has given me better results than any "secret sauce" will. Good luck. Here:

The culture was put in fridge at T8 in May 2023, took out October 2023 and several transfers were made. I ran 2 56QT mono tubs, both had 4 quarts grain (WBS with some popcorn 5% and rye 5%) were spawned to 10 quarts substrate with 4 quarts of pseudocasing. The difference being one tub got just coir, verm; the second tub got coir, verm and 1/2 cup of dried manure. Substrate was pasteurized in jars on stovetop for 90+ min. I let the coir only tub go for 3 flushes, just to see max yield, and due to space issues. From that coir only tub, my largest fruit weighed 112.0 grams wet and dried to 8.3 grams (its a little to the lower right of the Sharpie past the open area). Everything was dried in a Nesco at 165 for 48 hours.
Coir only tub pic:

1. Coir only 56 quart PE tub. 4 quarts grain, 10 quarts substrate, 4 quarts pseudocasing. Days on grain: 33 days Days till first flush (blobs): 35 days First flush yield (blobs, dry): 80.0 grams Days till second flush (penis mushrooms): 14 days Second flush yield (mushrooms, dry): 129.0 grams Third flush yield: 33.0 grams Total yield: 242.0 grams Test results: Sample consists of 1.0 gram of blobs + 1.0 gram fruits. Psilocybin: 8.3 mg/g Psilocin: 0.7 mg/g No other compounds present. Total alkaloids: 9.0 mg/g
2. Manure added 56 quart PE tub. 4 quarts grain, 10 quarts substrate, 4 quarts pseudocasing. Days on grain: 34 days Days till first flush (blobs): 43 days First flush yield (blobs, dry): 69.0 grams Days till second flush (penis mushrooms): 11 days Second flush yield (mushrooms, dry): 122.0 grams Total yield: 191.0 grams Test results: Sample consists of 1.0 gram of blobs + 1.0 gram fruits. Psilocybin: 6.2 mg/g Psilocin: 1.3 mg/g No other compounds present. Total alkaloids: 7.5 mg/g

Okay, so this is an update 2/11/2026: I saved the largest fruit from the above coir-only PE tub that finished 2/2024. The fruit weighed 112.0 grams wet and 8.3 grams dry. Here is the specific mushroom in the tub before harvest: And here is how I stored it for the last 2 years. Heat sealed in quart jar with single 2 gram desiccant packet. It was left on a bookshelf in a room that would go between mid-low 60s F to low-mid 80s F. There was no direct sunlight in the room, just regular office light LED bulbs. Jar was never opened until day of testing:

Test Results: Norpsilocin: 0.243 mg/g Psilocin: 1.422 mg/g Psilocybin: 1.906 mg/g Total alkaloids per gram: 3.571 mg/g
Obviously, the overall potency dropped, but psilocin levels seemed to increase. It's possible that the previous sample had less psilocin due to it being half fruits, half blobs. The HPLC service I use has also updated their equipment and standards in the last 2 years, so it's possible the first results were not as accurate. The potency loss over the 2 years, as stored was ~60% which tracks with other results #8 P.E. SINGLE FRUIT, from the original post above. However, that drop in potency was over 1 year and this is over 2; it's possible that potency loss has somewhat plateaued at some point.
