Jars Check

Hey, hello Myconauts, just a little check... :smile:

I have made a new spawn jars, inoculated by little agar pieces pre-mixed inside, they are 7 days old and as you see, there is some mycelium growing inside. This is my first time doing it like this. My plan is to shake it at 20-30% colonized, what do you think about it?




I have also taped the lid around because I think the contaminated air is entering inside from there as well.

No, the jars are fully closed; I have just taped around the lid for extra safety.

That’s not good, they need to get some air, usually by making a small hole in the lid, 6-8 mm, and putting a filter over it like a SFD, tyvek or polyfil. SFDs will last a long time so if you can get some, use them.

There is a 1 cm hole on the lid with microtape on it.  :grin:

They need air, remove the tape. If you don't have lids with SFD's or equivalent for gas exchange, You need to have the lid loose during sterilisation and post inoculation. Pastywhyte's Easy Lid Tek

As I understand the only way contamination gets in through the threads is if you have the lid too tight during sterilisation that it pulls a vacuum, then when you start opening it will pull dirty air inside.

Not fully closed as we think of it then but alright :lol: Still, micropore probably won't cut it but I don't know. You should have SFD's on the lids.

Sorry but that's horrible advice. A 1/4 inch hole with 2 layers of micro pore tape is plenty

Sure, but why ever bother using SFD's then? I use three layers for my brf pucks and that has been fine. I just hope he has more than one layer.

People were using MP tape long before SFDs. Lots of different filter material has been used: MP tape, polyfill, tyvek envelopes, tyvek paint suits. People tend to use what they have access to.

Besides, taking a filter off to replace it, even a bad filter, just increases the chances of contamination since you have to expose sterile grains. SABs aren't perfect, and not everyone has a FFU/LFH or knows how to use one properly. I'd take a bad filter put through a PC cycle over an unsterilized superior one any day of the week (then I'd run the superior one on my next round)

Fair enough, I retract my statement. Sorry, I have a bad habit of overstepping and I need to stop. I'm glad to be corrected when I'm being dumb. It's just that he said his jars were fully closed when I asked if they're getting air, that's why I said to remove the tape from his threads.

I think that my microtape around the lid is maybe unnecessary but not a problem. I'm always sterilizing my jars with a loose lid because I don't want pressure to crush my jars, but vacuum is another good reason that I didn't consider.

Does my jars getting enough air, I don't know, but I expected that it would get colonized much more in 7 days on temperature between 24-27 *C... maybe my mycelium got shocked when I have rotating and slightly shahing the jars to position my agar pieces lower in the jar, maybe agar is not cleanest or spores don't have best genes. Problem could be so deep, but I'm accepting the speed of colonization.

I'm not familiar with that term SFD, I'll check it out.

My biggest focus now is at what moment should I shake it and should  I shake it at all since I have mixing multiple agar pieces in my jars. I wonder what was purpose of mixing multiple agar pieces in my jars if I will shake the jars anyway... :confused:

Synthetic Filter Disc, some come as stickers and are thin, others are thich and will have to be glued on the lid with a high temperature silicone. Both are hydrophobic and doesn't get wet during a PC cycle.

I prefer the the thick ones.





You can shake them whenever you want, the point is to spread the colonized grains around so colonization happens from different parts of the jar and complete colonization faster.

I usually wait until I see at least 30-100 grains are colonized, but it doesn't matter. Then do another shake at around 80% and then one at 100% just to be sure I got to every grain and to see how fast they bounce back.

And of course you can just let them do their thing and not shake at all, it's up to you :rockon:

Oh, now I see. I have that Synthetic Filter Sticker; every one of my jars has it on. :grin:

I'm going to shake it at 25% because I want to shake it only once or twice to speed things up. I think every shake is a stress and shock, and then colonization in the substrate could be slower.
