What do you think about my new Agar plates, 3rd generation? Are they pure? Should I go for the 4th generation?

I'm going to refrigerate it since it is 80% colonized.
I have taped it with micropore tape; I think it is better than plastic film, because in the past I had moisture in my plates, and this tape will allow it to breathe some air.

"pure" is a difficult and maybe irrelevant concept IMO, and one which conflates a few different things:

* Clean: growing only what you want, nothing you don't.
* Uniform: growth is even without distinct segments
* Vigorous: is growth fast, rhysomorphic, or whatever other thing you care about?
* Isolated: here be dragons lol. Are you growing a single dikaryotic pair / one genetic organism? Not worth worrying about in 99% of cases.

Though it's a bit hard to see, those plates look clean and uniform, You could send them to grain. As for further transfers, it depends on what you want. If your goal is to get something you can safely send to grain and produce mushrooms, you're there. If your goal is to strengthen the culture on agar, that growth is a bit wispy/thin and could be improved, if that's something you care about. But there are many paths to arrive at a strong culture, and chasing vigor over many transfers can be tedious compared to growing them out and cloning a strong fruit, or germinating new spores and grabbing a sector that's healthy from the outset.

Also, plate performance is not a guaranteed indicator of performance in other areas. Plenty of things will wake up on grain, or put out some great flushes without ever throwing IG-worthy ropes.

Yeah, pure is an unreal term.

I'll use those last plates for inoculating. The logical next step for me is to develop a Vigorous rhizomorphic culture for future grain.
I'll have to wait for some good fruits to clone; chasing vigor over many transfers seems wasteful without experience; I'll take some lessons about it.

At the end of the day, it's better to use agar that is just clean than a spore syringe. Fruits from my current cake are so thin, I've been using a spore syringe for its spawn.
