Frozen straw and microorganisms
    #2932113 - 07/27/04 12:45 PM (12 years, 4 months ago)

Odd question here...

I pasteurized a large amount of straw about a month ago. Soon after, I realized that I did not have enough spawn to make use of it. It had drained/cooled for nearly 2 days before I simply placed it in a trashbag inside the freezer.

Now, microorganisms CAN live at these freezing temperatures right?... albeit at a severely hampered metabolic rate, yes?

If so, then the straw should be ok to use after thawing (as the only thing living in it would be the GOOD microorganisms). If not, it would require a repasteurization.
---

I thinking the first scenario is true, as bacterial and fungal movement in a freezer should be next to nothing. However, I think I'm going to repasteurize it anyhow.

Thoughts?

This is a strange question. I wouldn't be concerned with breeding microorganisms in a freezer, but you might have killed a lot of the thermophiles. I guess you could do it either way, but I have no idea of the outcome. With proper sterile technique and plenty of spawn, though, you should really be fine.

If it were me, I'd get new straw and pasteurize it -- it's really cheap and not worth messing up a whole experiment. It also matters a bit where you live -- I know that there is temp-resistant trichoderma that grows in Alaska....

Well I'm in the southern states...

I'd really rather not use more of my straw reserves since I haven't found a good way to cut it up (scissors are sooo tedious...)

I thought about leaving it outside a few days to let natural micros regenerate, then pasteurize again. Most of the jars I'll be using have very minor infections, so I'm not terribly hopeful here.

Does anyone have any obvious warnings though? Would it be horrible to simply thaw and use? Thaw and pasteurize?

Freezing expressed coffee grinds for long periods before innoculating with oysters has always worked well. No contams. I think you are good to go. Thaw and use. especially if you are using an isolate.

you can pick up a paper slicer fairly cheap at office depot or staples, unbolt the little guide bar and you have a nice little straw guillotine... watch the fingers
