What does mycelium grown in honey water look like?
    #2929140 - 07/26/04 05:20 PM (12 years, 9 months ago)

Does it look like little balls of cotton? What should I see? Could other white molds grow in honeywater?

Doesn't shaking help the growth in honeywater? More spread out and linear.

Shaking will help to dissolve air, which the mycelium needs.

Gray growth could very well be a contam, also healthy cultures have the Myc. floating in the middle contams are usually on the bottom.

I recently made four jars of karo cultures, two of them have big cottony clumps, and the other two have only a milky look and a few isolated threadlike strands. the milky stuff swirls when jar is agitated, so i was wondering about that myself. But I have seen pictures in documented teks with what looks like big clouds of mycelium, so im a little confused.

I recently found out that myc. grows in a few types, 3 I think. Anyway I would think some jars would look different then others. I have only used the honey tek 3 times and all three times it looked a little different. But I did not have any problems with contms when I used it ,so I must of had it right. Don't know what to say really haven't screwed with to much to know for sure.

i'm glad you asked as i am testing this very question. so far i have kept the same culture viable since march of 2003... i suspect it will be a long while before i find the EOL.

the culture lives in the .5 pint jar i initally started it in. viability testing is performed every 60 days by injecting a syringe full of sterile water then refilling. the resulting culture syringe is then used to innoculate 4 agar dishes. the resulting agar is used in wbs to verify the culture's ability to still fruit.

note that the original soultion ahs never been supplemented with any further nutrients. also, the culture stays refridgerated at ~35F.

A year is quite long.
What strain is it?

96ml tap water + 4g sugars of honey (8g of this particular honey = 4g sugars)

i think it worth mentioning that i stopped shaking the jar (has marbles in there) after day 3 of incubation. since then it has lived in the refridgerator.

the purpose of the experiment is to determine how long it would take under specific conditions until the mycelia was incapable of further growth and/or fruiting. I'm a bit disappointed that I didn't start similar cultures and treat them differently (ie, shake every 3rd day, store at room temp, etc.). I would suspect that any other conditions would lessen mycelial longevity.

How little spore solution can be used? [Re: 4hodmt]
    #2962432 - 08/04/04 01:04 AM (12 years, 9 months ago)

I innoculated 3 honey jars with:
1 drop
5 drops
10 drops

Has anybody had success with this little of an amount? I use 3mm syringes (hey they're free) and I am trying to keep a strain without exhausting my supply completely.

I just got some more malt extract today too, I think I'll try a couple jars with that too, but I need some guidance. What is the smallest amount of spore solution that anybody has pulled off.

Just for the record: I know this batch is viable, but the spores are barely visible when shaken.

Can somebody explain why you would use honey rather than a normal type of liquid media?

People can't seriously be this lazy, can they?
