Syringe germinating nothing but bacteria
    #2903029 - 07/18/04 06:06 PM (12 years, 4 months ago)

I have a light pan cyan syringe that I innoculated a few jars with also tried germinating on agar.. both times nothing but bacterial contamination came. On the agar I'm thinking it may have just been too wet (too many drops from the syringe), although it was on antibiotic agar. I'm gonna try again with agar and see what I can do since this a strain I haven't had the chance to work with yet, but if bacteria comes again on the next try, is there a way to salvage it?

--------------------
Looking for:
Ps. weilii, Ps. samuiensis, Ps. hispanica, Pan. cyanescens (any strain I don't have)

Have:
Ps. cubensis, Ps. tampanensis, Pan. cyanescens (Australian, Tai-KS, Hawaiian, Jamaican, Ban Lipa Noi)

PM for a trade.

Yes, I do all the work in a flowhood.

The agar and jars were both pressure cooked ofcourse. My hands are covered with latex gloves sprayed down with rubbing alcohol. I am using sterile procedures, I have just asked this question because the syringe itself may be bacterially contaminated and I was wondering if there was a way to salvage the spores.

It may take several plates to get rid of all the bacteria. Awhile ago I was working with a bacterial contaminated print, some of the cultures took three antibiotic plates before they were clean.

Have you tried antibiotic in the syringe itsself? Have you tried increasing the % of antibiotic? If you are getting germination, but are having isolation problems, then I recommend this:

I also suggest you lower your nutrient content, this may help as well...

Thanks!!!

I am just happy to contribute. It is much easier to describe something when you illustrate it. I figured somebody should do it, and I had the perfect opportunity. I hope somebody will find it useful...

Also lower your incubation temps. I've seen grain jars literally just soggy with wet spot that have recovered when placed in a low temperature area. For cubensis, 60's and low 70's has helped drastically reduce the ability of bacteria to overpower the mycelium, I'm not sure if it would be much different for pans.

If you can find one this time of the year, try incubating in a place with a temp of about 65. Mycelium will take a long time, but bacteria might take longer.

PM me with any cultivation questions.

I just looked at my profile and realized I had a website at one point in time on geocities, it's not there anymore and I have no idea what I had on it. Anybody remember my website from several years aga? PM if so please.

Damn.. still germinating bacteria all over the agar. I'll try a higher concentration of antibiotic maybe... really don't want to lose this strain.
