Goodmorning peeps. So you guys have convinced me to start testing my live cultures. My last grow did have a contamination issue, it showed its face in my monotubs. 2 out of 3 monotubs were taken over when the two tubs were colonized right before hyphal nots, really when the 2 monotubs were fully colonized. I don't understand how that happened after growing so good in the bag. Anyway, I purchased some grow jars, never used them before, but I will have the jars so I will take you guys advice and start making my own grow jars, so I will be in control of how everything is done, not a source that who knows their sterile protocol. I still not 💯% on making my own jars, considering I have never made my own jars, I definitely a rookie and will need some guidance, I want to be successful!! I have to purchase a pressure cooker, not sure on what brand to purchase or how many quart size to get!!
So at the moment I am about to order supplies for agar testing my live cultures!!
If I leave something out of my supplies list please enlighten me.
Pre poured agar plates. What kind do you suggest?
Innoculateing loop Alcohol burner- I'm not sure why you need this when you can use a lighter to sterilize the Innoculateing loop.I guess that's all I need, correct????
So I am sure there is a post explaining exactly the sterile procedure to do the agar test "sterile free "..
If you guys will please tell me exactly how to do the live culture test, I would really appreciate it!!!!
1 question I have for sure is:: so the Innoculateing procedure is when I have a knew live culture syringe that is in the package and sterile, and a crucial step to be contaminate free,
How do you open the live culture package, take out the syringe without the needle, take the plastic cap of of the live culture syringe, get your sample out for testing and repackage everything in a sterile fashion to use when the sample on agar plate is in and its time to inoculate the jars of grain. How do you do this and keep everything sterile?
Remember I have never tested live culture before and I am clueless on how to do this test!
So how much of the live culture do I remove from the syringe for testing on agar?
How do I put the sample onto the sterile pre poured agar plate?
What next, I assume I wait for results from the test on my agar plate.
How long does it take to get results from the agar plate?
What am I looking for? What does a good agar plate, a agar plate showing my live culture is good!
What does a agar plate look like on if there is a problem with my live culture?
I assume if there is contamination in the sample of live culture, then the live culture syringe is no good and do not use it.
If you guys purchase a live culture and it comes up showing contamination in it, do you return it to the source for a replacement?
Do the sources where you purchase the live culture from, do they normally send you a replacement syringe free of charge or do you just lose your money?
Seems to me, if they want to keep you satisfied and keep your business they should give you a replacement!!

I just want to be successful and I am taking your advice and start testing my live cultures and making my own grain jars. I do need a pressure cooker, so I will purchase that immediately. What brand and how many quartes
I apologize for this post being lengthy. I just really need your advice. I will be ready to inoculate my jars very soon, as soon as I order and receive the items you guys suggest I order for doing my agar testing.
