If you put your LC to agar with success, I’d take an agar transfer to your new broth, not inject the vendor bought LC to broth.
Either the LC is dirty or your sterile procedure is bad. Either way, agar is going to help you out

Spores to agar to more agar until clean, then to grain spawn, g2g, storing culture for later, g2g , spawn 1:3 mixture of just about anything, straw and composted manure home depot brand, oven pasteurization the easiest way, unless your doing less then Frank's pasteurization tek. And case with I'm finding out cactus soil same pasteurization as top.

Why cant you conveniently use manure? Its real simple. Are you just worried about smell?
recipe I would use:
30% Coir / 30% Verm / 20% Straw / 20% Grain (millet or rye)
Use that as your core. Maybe even fluff the verm with BRF.
Use the EF-Tek for reference and Ideas

Quick question about hole placement for these tubs.
The plan, I have 2 x 90L(95 Quart) Tubs, and they fit 4 x 5L (About 5.3 Quart) Tubs inside them as in picture.
The 5L Tubs are 140mm(5.5 Inch) Tall, I was going to fill them to about 1 inch below the top of the 5L tub.
I planned to use the Pastywhyte’s Ez dialed monotubs tek for hole configuration, and he says to place holes 1.5 to 2 inches above the substrate.
Since I plan to fill the 5L tubs to 1 inch below the rim, I would assume to place holes in my 90L tub about 1 inch above the rim of the 5L tub giving me 2 inch clearance from substrate to holes.
Do you think this would be correct?
Final part of the question, since these are 90L Tubs and Pasty's guide uses 66 Quart tubs which are almost 1/3rd smaller, do you think the 3 holes long side and 1 hole centre short side at substrate level will suffice or being bigger tubs would you be tempted to add another hole to ethier the long or short side or both?
Thank you.

I would start with less holes and add more if you need

Mackload said: This is almost two months growth. I used some of the agar and left the rest at room temp. There are some little discolored bumps, not sure if they're maybe metabolites, or tiny mushrooms? Might hard to tell from the pics, condensed lids. I don't want to open them for a picture.

Looks a little wet for just condensation .. and looks a little backyard. Pic from under neath with light on top?
pic is from on top but with a flashlight under 1,2 and 4. Daylight pics will be better
Have you looked from the bottom? When Condensation is unbearable, it may help to notice any peculiarities or discoloration.
Also, may I suggest pulling the cling wrap a bit more when wrapping to make tight

I dont have the supplies to make culture slants, but i have prepared agar. I made a transfer yesterday that I would like to store in the fridge for as long as possible.
Is it better to let it colonize most of the plate before storing in the fridge, or is it ok to put in the fridge as soon as the transfer is done?

Best is subjective. That being said, I like to give the mycelium a few days at room temperature to jump off of the wedge and begin colonizing the new agar before storing agar cultures in the fridge.

I take meds that suppress my immune system so my doc told me no manure in my grows. So I use liquid plant fertilizers. Way more expensive than using manure but it’s quite convenient.

I don't use straight grain spawn with Pans and a few other people who have grown them awhile don't either, as getting them to run on grains can be hit or miss.
I use a mix of substrate and grains (if you can't use compost, do straw and coir, the last jars I ran are coir/hemp straw/manure with 1:4 grains all sterilized in the jar).
Home Depot sells Black Kow which is compost from cow manure. It's very well composted and practically soil. I've grown plenty of Pans on it.
Leaf Compost looks like another option. I haven't fruited this yet, but it's running well in jars. I bought a big bag of it from the hardware store.
Here's my various experiments along with a few other people that have posted in my journal. Soon I'll put together a proper tech with different recipes for people to follow.
Anyway here's that thread if you want to take a look.

Well to be fair, I asked the forum and you guys gave me a resounding “ask your doctor.” So I did. From her perspective the issue is handling it prior to pasteurization. I trust her & the ferts work so I’m not trippin

Wiping your own ass exposes you to more dangerous fecal matter and microbes than processed manure, but okay doc.
I'd just wear gloves and a mask and go for it
