LMEA plates and the addition of yeast 2

As an experiment I have altered my LMEA plate recipe to include yeast at 2 concentrations and the results are quite improved from the formula without yeast.
However, there is a significant loss of clarity with the addition of yeast that I am unhappy with. Is there a better way to fully dissolve yeast?
These 3 plates all have 2% agar as the gel agent, note the difference in clarity of the LME only vs the lack of clarity when adding yeast. However the difference in growth is significant enough to be in favor of yeast.
Note that while the yeast plates are 1% lme compared to 2% lme in the yeast plates, the growth of the 1% is consistent with other 2% lme plates I’ve poured, I was just testing 1% in general to see how much more clarity it offered compared to 2%. In retrospect I should’ve also made a 2% as part of this experiment for comparison.
The first of each color/clarity of plate was taken after plates had cooled and are just to show the composition. The second two photos in each color were taken 5 days after a punch from a MSS to Agar (2% agar, 2% lme) plate was transferred: Orange is 1% LME and 0% yeast

Pink is 2% LME and .1% yeast

Purple is 2% LME and .2% yeast

Traditionally yeast extract is used as a media ingredient.

Update, Day 11 after transfer. .2% yeast is still the leader and the .1% yeast seems to have altered its growth to an outer ring of lighter growth. Perhaps running out of nutrients? Curious to see is he purple plate does this at some point.

Idk if anyone is following this, but I ended up transferring these t1 plates as well as 3 others t1 plates I made from a very bacterial MSS to t2 plates…
![]()
But…. at the encouragement of my mushroom boys, sent the t1 .2 yeast (purple plate) directly to grain!!!
 Here’s that plate:

I chickened out a teensy bit and split the plate between 2 jars instead of the 4 I had originally planned. Fingers crossed!

Personally, I have had a bad experience when adding "brewer's yeast extract", when transferring my plates a robust and rhisomorphic growth was seen, however these plates, at the time of fruiting, had low yield and rotted quickly, showing bacteria in it. Anyway, your thread is quite promising, I would like to see your results. At this moment I am collecting more data and in the coming weeks I will make a more detailed publication on this case, it could be of great help to the community

You have low yield because your tubs are not happy whatsoever, they are dry AF and fighting massive contaminations as apparent by all the dried metabolites on your sub surface. I highly doubt that was caused by adding yeast to your agar.

What would you recommend in this case? I live in a fairly dry area, however the controlled humidity is in good ranges, the substrates have a sterilization time of 45 minutes and the wheat approximately 90 minutes. Regarding the ventilation ports, they are enough, should I reduce them? Soon I will have plates without yeast, only potato, agar and dextrose. I will record the difference in the crops, generally albino ones tend to fruit like in your fixed image quite well Greetings, any suggestions I am willing to listen to.

Are you using a heart shaped leather tool as an agar punch?

Now now Meta, play nicely…Let’s be sure we are using kind words when we approach constructive criticism.
