Funny how it's possible to tell you're in the southern hemisphere just by seeing the tiny pseudo-stones in your substrate jars. I've only ever seen these in warmer months of the year.

I didn't notice any pseudo stones in the jars. I had PF-Tek substrate on top of the grain, maybe you are referring to the vermiculite? Could be bits of BRF too.

Ug. I feel like pounding my head on my desk... Everyone keeps talking about BVI growing really fast, but mine doesn't seem to be doing much. Any advice?
I swabbed 2 plates using a (purchased) spore print on 2/6/24. Here's what the plates look like today:

You can see in the right plate where I pulled 2 plugs for t1s on 2/11, photo today:

Is this just all mold?

Ah yes now that you mention it it's probably verm But it does look a bit like stones that they can make if it's warm.

0.25% dextrose, 0.25% soy peptone, 2% agar - it's what I had handy in the lab at the time. The growth looks similar to what I saw on GWA recently, but I don't have photos of that.

Could be the recipe. I'd change it back to GWA diluting one part of grain water with 3 parts of water (could even go lower I suppose but I am not doing much GWA) or MEA at 0.5-1%.

I’ve had the same plates like you before. Unfortunately I can’t remember what was the problem. It is either a heat or moisture problem. It is definitely not a bad print. Mine had a excess moisture, but yours looks good to me so I’ll bet on improper temperature. How do you store your plates ?
 Here is the only photo I have. In the center it looks bad,but it grew normally after I put it in the right conditions.

Ok, I'll try diluted GWA and go get some LME... I'll throw everything in there.

This is from a print Treefrog gave me on lmea. I would definitely try a different agar recipe.

1) Sanity check: MS > T1 / T2 > grain > grow > clone > clean > T1 / T2 > back to grain to confirm. No need for isolating sectors until a decent F2 grow results?
2) Also, I'm moving soon and will have more room. If I care to do it, is a tent setup the most go do for pans now-a-days? I can do either tent or monos although I do enjoy building stuff so it's no big deal to get a homemade tent up and running. I've transitioned to pans only based on personal and local feedback.

That’s why I keep saying there’s a clear difference between ttbvi and bvi! The bvi culture I have is SLOW. The ttbvi I have is FAST

I think it's more domesticated because Gordo has been domesticating it.
Like the original wild prints that became B+ may have been P. Cube Florida strain (making this up as an illustration).
Pan. Cyan TTBVI seems like it could be the B+ of the Pan Cyan BVI strain.
My understanding though is that both the BVI and the TTBVI floating around came from the same guy in the British Virgin Islands. And reading through some of the comments of him not appreciating all the attention from supporters on Gordo's Patreon, I'm pretty sure he's a real person.
To add to the mess, if you got prints from me labeled BVI, they're TTBVI. I just hadn't sorted this out in my head yet, and I think more precise language is better, so I'll be labeling them as such in the future.
Gordo's prints, via Supa's culture, helped me uncover 40-year-old trauma. Whatever the truth, I'm off the Gordo hate train. He seems very generous and sure, he's marketing himself on Patreon, but I can't hate someone just for marketing in our fucked up capitalist world. I do think he should have gotten consent before outing the original collector. But again, consent is something we're learning as a culture. And it seemed that Gordo's intent was for his Patreon's to support the guy. (But he didn't want the attention, as I pointed out above).
I think Baba's more on the right track with that, monos are tried and true and cheap. But that's going to take time, especially if we're still dealing with prohibition. Because it is taking a lot of culture work. Eventually, I hope we have a Pan Cyan Rainbow Yogi Cum strain that has been domesticated for monos, so that these fruits can be more accessible to more people.

My advice is to keep those plates someplace a bit warmer, BVI is a tropical strain. TTBVI is a more domesticated child of that strain, imo, that's likely running better at room temperature due to genetics and epigenetics.

Working "Mini JCM Aqua Tubs":
- Inside, floating shoebox w/ ttBVI.
- water temp in low 80fs / surface temp of the substrate holding steady at 75f.
- Other items pictured: QT jar for "pan-cakes" and agar dish w/ 3 grains pulled at S2B -> already showing strong growth off the grain onto agar, will be doing transfers in next 24 hours.

Also did up 2 more jars following Tree Frog's "Pan Poo Sub"
- Went w/ drier sub as I'm using LC: I've always had challenges getting saturation correct when using LC, often too wet. Also, not sure how well Pan LC does, but the culture looks very strong on agar and in LC mix.
- The first round of jars is nearing completion (agar wedges used), both currently on day 14, which for me & my environs is fast
- Also went w/ just rye berries instead of millet/ryeB mix
New Jars: Pan v PHV LC

Round 1 jars:(hard to see w/ lighting and sub in jar - nearing completion)
