Hi everyone! Here is a little write up to describe a little tool I made to transfer really small agar samples (can be useful to have micro samples to select a sector, a clean zone or even using smaller plates...) for agar to agar or agar to small LC it’s relatively easy to make and to use:
Can be sterilized in PC & needles can be flame sterilized during a transfer session (just let it cool before taking next sample)
Can be used in very small or deep plates even probably in a slant..
As the sample and the contact size of the tool is small I think it reduce risk of contam (so far I got good results with it even without a flowhood)
As the sample is very small it can help to do precise sector isolations (so potentially reduce number of transfers needed).
Also can be useful for taking a very localized clean spot in a dirty plate.
It’s not difficult technically to practice & it’s relatively quick to use with not a lot of movements...



Materials needed are easy to find and cheap, maybe most of us already have those on hand, basically need:
a syringe, 2 syringes needles with one thinner (enough to pass trough the larger one) & longer (to make it longer I stick a fine sewing needle that fit at the top of it..) and the other syringe needle larger & shorter (cut), and a spring.



The tip of the larger needle is cut to be flat like a little tube to take the agar sample & a bit bended because if it’s straight the samples will stick to the plate most of the time, if it’s bended after the punch you just need to turn the syringe (1/4 to 1/2 turn) before removing and like this the sample stay in the needle that’s the trick! and after you just need to push on the syringe to transfer the micro-agar sample to a new plate or a LC.

Thanks! I think the pink ones are 18g but important is the size of the thinner one compared to the larger one to pass trough it..

I didn’t check exactly, I should do it with a little timer I would say around 2min, I usually flame it directly after the transfer so during labeling and wrapping time it cool down for the next use and if you work really fast why not build 2 to have one cooling when using the other..

As some people here and elsewhere asked what Gauges needles i used I searched for the color code (I ask syringes by colors in the pharmacy) so if we refer to that code should be: larger one pink 18G 1,2mm and the thinner green 21G 0,8mm



(Not all syringes companies use this code)

Sorry if this is a dumb question or if I missed it but how did you get the screw into the plunger like that?

I'm rigging together one of these the only issues I'm having is how to get a screw in there like yours and I can't find a decent spring or needle but I've got them lying around somewhere

Edit: nevermind I popped the rubber plug off of one of my syringes. A few syringes I tried the rubber would not come off

How did you end up cutting the syringe needles without mangling them or crimping them? Did you saw em?

I tried cutting one without really thinking about it and just crimped it closed after it was cut, I kinda corrected for it but now the needle is really short. It works but it's shorter than I'd like it to be

I have a few more needles to tinker around with but I don't wanna just destroy em if there's a good way to cut em down

It was crimped after cutting it with cutting pliers but just reopened the hole with a knife or something like that and then used a stone sharpened as finishing to have a clean shape..

i have found puncture needle that have an obturator inside
so with a small spring and a 5 ml luer lock syringe i could make something like your tool just by cutting with a dremel the needle by 1 cm

i just want to see if everything will be ok after PC it

I’ve got a simple setup. 4 inch 14 gauge dispensing needle with an 18 gauge piece of florist wire attached to the plunger. I cut the needles down with a dremel and sharpen them with a bit of sandpaper. 2 to 4 inches in 1/2 inch increments.
