I try to always take from the leading edge, or as close to it as I can get.  I never take from the middle, that's what you're trying to transfer away from.

How long has the plate been fully colonized like that? Do you want to wait for pins to form or run a tester?

Oh, yes for sure. I was talking about selecting mycelium for grains, not for transfers.

It's been like that for a day and a half. It grew really fast.

I've already made a transfer to a new plate, now I'm contemplating what to do with the existing plate. As the mycelium is so vigorous, I was thinking of inoculating a couple of jars with it and see what happens. Just wasn't sure if I should discard the middle or keep it ...

I have two such plates. Almost exactly looking. Not quite sure about letting a pin form on the plate. I'm new to agar and have to research what are the benefits of using an in vitro pin, why you'd wanna do it.

I guess my Sunday school teacher was wrong. Sometimes Force IS the answer.

Got impatient to try my new glass plates and poured way too hot. Nothing was working to clear them until I started standing them up in the forced air of the flow.

You shouldn't if your culture is clean and ready for grain,then you can cut up the entire plate.

Given that it was a t1 with a slightly questionable middle, I would absolutely transfer from the edge.

Hi


This is my first time trying to do anything with agar
First up I have a clone attempt of a pearl oyster. I took three pieces and placed them pretty haphazardly on the plate. I figured there'd definitely be contamination in there somewhere.


Is this sludge that's surrounding the pieces contam?


How would I go about isolating the pieces that seem to have mycelium growing out of them? Do I wait until it grows over the sludge or do I try to chop a bit off of the one that's vertical in the top right?


Also I got a pink oyster liquid culture and read that you should test it first on agar so I squirted a couple drops onto this plate


Is it normal for the right section to be so condensed and small in comparison to the left section?


This is the first plate I squirted some pink oyster liquid culture onto lmao
The plunger was a bit stiff so I ended up squirting a good bit onto the plate on accident oops
It's been about a day since I took this picture and the plate is a little further along but generally looks the same. Should I try to isolate parts that seem to be growing faster now or wait a while longer?

Thank you for your time and sorry for all the questions I'm very intrigued and not sure how long I should be leaving these to incubate in general before attempting to do anything with them

Transfer the plate pins to new plates.

Do whatever you want with the original plate.

It appears that you just inoculated those plates 2 and 5 days ago.  Try and be patient and don't look or touch them for at least 10 days.  Just pulling the plates out into the air/light (even if they are wrapped) can and will cause growth stunts on the plate.
Yes that white sludge looking stuff is contam.  Just throw it away.  Agar is cheap.  On the plate you applied too much LC,  Yes, just experiment with some transfers. Try to take specimans just from the edges.  One small piece for each plate at first.  By small, I mean the size of a #2 pencil eraser. Just a sliver though.  Good luck Lemon

I didn't know that at all I've just left them on my shelf and look at them every now and then

I'll be more patient and let them do their thing before jostling them around more

Sounds about right yeah. I think I'll try a transfer just to see firsthand how it's gonna go wrong so I can understand it better and get that experience
