Some anonymous person on the street corner stopped me and asked:

"Is it okay to cut out visual contam from the jars and spawn to coir? It looks like 2/6 jars have a small contam...can throw it away if that's what is needed, don't want to contaminate other totes etc"

It’s basically a lost cause. It could have contamination anywhere you’re just able to see some of it. The jar is dead. The most you could do and I was able to do this. Take a chunk out of the jar that looks like the most viable bit of mycelium. And then you can transfer that to agar and clean it up.

At least then, the spore can be salvaged.

I still have some mycellium on agar, and other jars going.....I was just excited to start a mono with 6 jars full!

I’ve got a shoebox and a sweater box of starry night APEs. They are looking pretty blobby on this first flush , while I had some other first flushes from the same culture that were great. I am thinking about harvesting tomorrow and hoping for a better shaped flush 2. I was out of town for the past 5-6 days so I haven’t been able to watch the blobs to see if they are still growing in the past few days. Does it make sense to harvest as is or should I observe for a day or two and see if any growth is still happening? Anything I can do between flushes to reduce chance of blobs?

Never grew APEs, but from what I've read, it is usually the second flush that blobs out. I would expect the second flush to be different. Then again, I am a known optimist.

edit. Still not sure if I am right, but I meant to say that it is the first flush. Damn, I should double check my posts.

I am a bit confused about genetic diversity and isolation and gene stability in cultures vs spores.

I have grown some starry night APEs, and their fruits thus far do not have a starry cap. Do I keep looking for a great specimen and then clone it? I started from a culture I bought so I don’t know why it wouldn’t have the desired characteristics.

Second question… SNAPES and other varieties that don’t have heavy sporulation, swabs are a great way to get the spores and share them. But if I am trying to share swabs of SNAPE in the marketplace, should i wait until I get a perfect specimen? I understand how the genetics can be isolated through agar transfers and cloning, but I don’t understand how the diversity gets back in the spores after getting a culture to a monoculture or some selected gene characteristics

SNAPE works much better as live culture because the stability of the specific genetic trait that brings out the starry caps is unreliable.
can work from spores but not necessarily.

what was posted above that is wrong info.

(edit: finally i think it could’ve been a typo)

APE often produces blobs on the first flush (es) because of  the overly nutritious mix. later flushes produce more normal looking fruit.
high bulk ratios at spawn can help. try 1:4 or higher.

casing can also help, but imo it is most related to overly nutritious mixes

Does this imply you could use half a qt of spawn for a shoebox with 2 qts of coir for better results?

As opposed to 1 qt spawn 4 qts coir which would seem risky if your spawn wasn't 100% perfect but still good enough to expect fruits?

Those are both 1:4 ratios.

You could split the jar if you wanted.  But, if the jar was contaminated you'd end up with two contaminated shoeboxes.

That said, I use 3/4 quart jars often when looking for genetics and basically do what you're asking.  Just do it in separate jars.

Some very beautiful molds, but mold nonetheless.

My question is how fucked are the rest of my plates if they were chilling below that in the same sleeve?

I keep plates in plastic sandwich baggies instead of wrapping them and I’ve seen one plate contaminate and the others sit in the same baggie for weeks and not contaminate. I’d say quarantine that sleeve for an extra time period to ensure that they are clean, but my gut feeling is that you’re probably okay. Depends if the nasties are in your sleeve itself or if that plate was the only thing that got contam in it

They contaminated before or during being wrapped. If the other plates haven’t contaminated, then they’re fine.
