Petri dish vs ketchup cups for agar
    #28667462 - 02/19/24 09:58 PM (4 months, 4 days ago)

Will ketchup cups works fine for agar or are petri dishes necessary?

A lot of people have been having great success hot pouring into condiment cups lately.

Traditional petris are not required.

Anything that should be done differently if using condiment cups?

As soon as you put the lid on start stacking them.  That will reduce condensation. Also, try to give yourself a 1-2week lead time. This will further reduce condensation and reveal any contaminated cups. Upsides are no wrapping, easy to tiger-drop, and disposable/ recyclable.

What do you mean by lead time? And do you ever have trouble seeing if there’s contaminants through the lids?

Prepare the cups in advance so that you can see if your pour was clean and to allow condensation to naturally absorb back into to the agar.

I no pour 'slime cups'.  I've ran them through several cycles.  Similar to ketchup cups but a bit bigger and the lids are pp5.

After they run and before they cool I transfer them to lunch bags to reduce condensation.  The top few will still have some but it usually clears up.

I sometimes get pools of water on the top plates because of the condensation clearing on these top plates.  But I just dump it on my work surface before doing a transfer into those ones.

cut your petris upside down and use the tiger drop. cut into 9 pieces in a a tic-tac-toe pattern and close back up. squeeze around the edges a lil bit and carefully inoc your chosen vessel.I usually place the cup right next to the lid, which is already loosened to where it lifts right off. i set the lif of the petri atop the jar lid and swiftly position and squeeze/tilt.
This is just for inoculation; there are  a plehtora of procedures you can use to enhance your procedure with cups. I am working on a write-up with gifs for instructionals on preparation and placement, pouring methodology, resting (optional), transfer, cloning and inoculation in several styles.
good luck with your cups man ive been using them for years and they have never failed me. The only thing to be careful about: make sure the lids you buy match the cups! make sure they are sterile pp5 they should come in a sleeve and are for kitchen supply.

The main problem with the cups is they’re so small you can’t really let the myc grow out enough to get a good look at the growth and sectors. I started using petris for germinating spores/cloning/t1s and then switch it over to cups once I’ve got a good culture going.

I use to use condiment cups, they work fantastic, but as AKA has stated they are small, the only reason I switched back to petri dishes is the height of them made them difficult for me to use.
If you plan on using them, I have found some cups that are 3.25oz, a standard cup most restaurants use are 2oz.
The 3.25oz cups are the same height as 2oz cups but are bigger around. If you PM me I will share the ink to where I get them and what I get.
As for pouring them, I do 10 in a stack, then when I have a stack done I take an empty cup and fill with hot water and place it upsidedown on the top of the satck, this way the top heated cup is the same size as the lid, let it cool for about 20mins and you will have little to no condensation.
To help aid in stacking more than 10 cups I made this rack to hold all the cups exactly centered upon each other and allow me to stack them 15 high.

this is the final product

I use ziploc twist mini 118ml for T2, T3 transfers. They are good for no-pour because of the twist lock mechanism(no modifications, no filters). They come with high walls which can add extra protection during transfers(good for tiger drop). By turning them upside down, water pools on lid farther away from agar's surface, so condensation is not a problem anymore.

The only issue is loss of visibility because of transparent blue lid, especially for the first transfer. For germination plates, I use Alien's Holy Grail No pour Agar unmodified containers..
