Flick the plate with your finger a few times and see if it turns green.

I did that. I'm not sure how quickly its supposed to turn green. It still looks the same. But it does look suspect right ? I'd be bummed if this syringe was a bust. I'm pretty excited for these. Tidal wave looks to be doing good though.

Couple hours for green to form if there were spores clinging ready to pop.

Parts look... disorganized to be sure and possibly discolored but some parts just look tomentose to me. I dunno. I've seen worse plates. I think it's worth transferring from the more uniform sectors.

Thanks for your input, as well as yours Fiddle.Im happy nothing turned green. IL definitely take some transfers later. Hopefully it grows out nicer. Will update in a couple weeks

How’s this millet spawn looking? Growth seems kinda weak/wispy overall, with little spurts of rhizo growth here and there - but it’s chugging along and doesn’t appear to have any wet/slimy spots.

Thanks!

As long as I’m here… Is this the dreaded Mycogone? Some other contaminant/stressor? Or just some funky blobby growth?

I'm guessing the top one is good while the bottom one is contaminated?
Do I have enough to start transferring to other agar plates and grain?

Could we get a better shot of the top one?

It's not really about quantity. You can transfer a tiny wedge to grain though bigger wedges gives more mycelium to spread around for more inoculation points.

It's about getting a clean culture. Not necessarily quantity.

When transferring a master grain jar to multiple other grain jars, what's the best way to avoid contamination?

Sanitize the outside of this grain jar. Put it inside of your SAB. Or in front of your LFH. Do the same for the jars you intend to inoculate. Then do it.
I have not heard of someone doing this for the record.

To me that appears to be mold but I'd wait for a more seasoned eye to confirm just cuz germ plates always give me a bit of trouble.

Loosen all jar lids, remove the lid from your master and set it down.  Open your first jar and hold the lid in your offhand.  Pick up master jar, roll the master jar gently back and forth to get the grains to pour out while holding it above your other jar.  Replace lid on receiving jar. Rinse, repeat.

There's probably videos online demonstrating the rolling motion.  It's a wrist thing.

I learned it from Stamets Gourmet and Medicinal Mushrooms.

It's not a great tek for SAB as the flowhood allows you to hold one jar over another without dropping crap and gives you more vertical space to actually do the pouring.

Also, I prefer 3/4 qt jars for masters as they're similar to pints in shape but just bigger.  The quart jars have that curve that catches grains.  But I've gotten comfortable using both.

It's either mold or the most disorganized growth I've ever seen. Unlikely.

Show the other one again? It looks like there might be since transferable growth if you hurry.

How do you get burned agar off of glass petri plates after oven sterilisation?

You soak the plates in hot soapy water.

Feel like I'm having bizarre issues with cult recently.

My agar has stayed liquid post sterilisation & cooling but with a tick layer of sediment at the bottom..

Is it possible my scale malfunctioned (or I malfunctioned) & I didn't add enough mix? I usually do 50g of premix (LME, Dextrose, Agar) to 500ml of water & have no issues, it's in a screw top wine bottle.

I'm going to add more agar mix & re-sterilise, I'm wondering if double sterilising will cause a problem?

50g for 500ml is actually a heavy mix.
I use 10ml agar, 10ml lme for 500ml water.

Perhaps I'm tripping. Few days of heavy drinking has gone to my noggin. I shall try again lol.

Derrayld & other tub users, when you buy fresh ones do you tend to wash them out first or not bother?

Just got a large haul & I'm debating whether it's a waste of time or not as it's of the essence today.

They look pristine the only concern is chemical residue from manufacture.

Cheers

I don't bother washing new tubs but I do drill holes and wipe it down to remove the excess plastic.
