Agar slants. Wood or no wood? * 1
    #28665243 - 02/18/24 09:48 AM (10 months, 23 days ago)

Just been looking into slants now I am getting better at isolating my strains. Just found a website that has the option of adding wood if you want. I presume this is because when we find fungi in the wild in the forest we see that some grow very well on trees? Do any of you use slants? And do any of you add wood? And if so, does it make any noticeable difference to your cultures?
I just want more experienced opinions from this large community 🍄

Slants are becoming an outdated thing.

Stasis storage is easier and better for the mycelium.

For sure.  Going to suggest it's added to the old information thread.

Just read. Recommend

I also read & like fathster dry method awhile back then(glad he bump it) in the thread., when I was still cultivating full throttle.

As well as small wedge in distilled water, haven't tried it as I was still afraid - sloppy back then.

Only manage to make few orissa agar slants with sticks. Before I threw the other cultures - moving to a new place. i think they almost 4yrs+ now(but unfortunately out fridge), Will try em out as time permits.

Small wedge in distilled water is what I use now.

Still in process transferring all my edibles off slants.

What is the method for sterilising paper strips using this technique? They will fall apart.
I was thinking maybe between two pieces of glass but I don't have any. And how could I stop it clumping together during the process? I am up for the challenge, but sterilising paper strips or paper about a cm across and the width of a petri dish doesn't sound easy.
Any suggestions or if you or anyone else has done this please suggest the method for successfully sterilising paper strips.

My bad.

I was linking the thread because the best ideas are in the replies.

Here's a simple one.  Take colonized grains and dump them in a sterile test tube or empty sterile container.  The grain will dry out and you can drop one later on a plate.

Mushboy has stored grains like this for over a year and revived them.  We're prodding him to test some three year old grain.

Another is to drop agar wedges in sterile water in a sterile container (a test tube works but there's smaller containers that make it easier, I use 0.8ml ones a friend gave me).
