Gamma irradiation 1

This is a subject I've seen discussed on a few older threads but... as I have a little left the a syringe and access to the equipment I thought I'd give it a go.
 This jar has been filled with standard 2:1:1 verm/brf/water and has been inoculated with 2ml from a spore syringe which is producing good results on other jars. However... in order to sterilize it, it has been subjected to around 35Gy of gamma radiation. What is odd is that unlike it's sister jars which were steamed for 90 minutes, this one has no growth whatsoever even after a month. And ideas why?

Yeah, that'll be the big green ring binder to my left... doh. The substrate is totally the same colour as when it went in. It was handled exactly the same as the steamed jars, which are now all full of lovely white mycelium.

With any industry if you need to ask "Why aren't people using this tool" it usually means:
It is too expensive for mass scale
It is not efficient at mass scale
It is not consistent enough
So that is probably why your jar is not performing as well as the favored preparation teks that are most used

Or someone doesn't feel like investing thousands/dozens of thousands just to test out if it'll work. Well, also gamma rays are dangerous. I believe it should work because gamma rays don't give a shit but something something depending on how you expose it and stuff you know how it goes.
Maybe it altered something in there that made it toxic to mycelium or your syringe was just toast. I believe it's worth trying again if it doesn't cost you anything. One sample is too easy to fuck up.
Now that I think of it, water is also a good insulator against radiations. Maybe you need to lower the water content and expose it to more. And also lead and some other metals which could be in the verm. Well, you should try with grains next time or something that you're sure will get permeated.

The calibration procedure for the equipment actually uses pint bags of water which the radiation goes straight through like it's not even there.
I think my next port of call is to give it another squirt from the syringe.

I mean it should work right? There's nothing in that jar with a nucleated cell that was left alive after the irradiation so maybe it's a little too good?

How deep does the radiation penetrate? I know the scalpels and some lab equipment we used are marked that they are sterilized with gama but those objects always seem really flat and thin without any bulk to them.
Are the rads going to penetrate the center of the jar the same as the stuff at the surface?

There will be a slight difference but the equipment is certified to deliver a minimum 35Gy dose to what essentially is a 300ml liquid pack. The indicator label turns at 25Gy. There's nothing really radio-opaque in the jar so I reckon the whole thing has received a dose of at least 30Gy.
