Ok, then the contamination makes sense. I assume you're cooling the blade on the left side of the plate. The contam seems to be on the side you're opening the most. I would try raising the lid only on the working side, just enough to work on the plate and see if that helps. It should force your hand to work beside the plate rather than above it. Also, try working a dry sab, no spraying at all.

Yes, I'm cooling the blade on the left side edge so only the tool is above the plate. I also hold the scalpel towards the "tail" end, so my hand won't come above the plate when placing the transfer in the receiving plate. I tend to open the plates and slide the lid up to halfway off, I guess I can flip the blade more sideways so it can fit through a smaller opening, or go extra slow and see if I can reduce the opening more without bumping into it. Thank you for the tips!

got lucky last year with MSS > grain. y'all encouraged me to dive into agar. BRF was all i had on hand, using 2% agar. thanks for any feedback.

I believe this is day 4 of the most recently inoculated plates and there's no growth of any kind which I think is a good sign, assuming something pops up in the next few days anyway..

A few days ago, I took a clone from a waterlogged fruit. It looks like all the red spots are likely to be bacteria. Does the green spot look like a good place to start transfers?

I tried to take a better picture from above, but the lid is cloudy with limescale deposits from the inside.

Quick noob question regarding BRF agar.
Is it advisable to use heat while mixing the ingredients or should i even pre-boil the mixture before pcing?

If you heat it before the PC cycle it'll give you a better idea of how thick it's going to end up being after it cools. I once made wheat flour "agar" and it turned into consistency of a brick after PC-ing

Hi all! I’ve got a few quick agar Qs from my first time working with it. The strain is machine elf, all the plates are T1. (I think I should be able to take the T2s to grain.) Here is the best plate of mine. I am planning to take many transfers from pretty much all around it. If that seems wrong to you, let me know!



Here are some other plates of mine, with less strong growth and some wispy stuff that slightly concerns me. Are you put off by the wispiness? Does it look like mycelium to you?


Finally, I was just wondering if y’all had any other comments/suggestion. I have been surprised to see the vertical growth some of my plates have done! I didn’t realize that would happen. Thank you!

If they're MS I'd recommend going further like T3 or T4 before going to grain. The first three pics that one looks ok. The vertical growth is a little differenty kinda. What percentage of nutrient are you using?
The other 2 wispy ones I'm not a fan of they don't kook right.

Yes that would be MS(multi spore) That's a good recipe and tek. I'd do more transfers and see what happens.

Pretty. Do you use a scalpel? I’ve seen scalpels with detachable tips. But I feel like I could just boil a random butter knife, and that would be enough if I brought it into the SAB hot.
Am I wrong?

Maybe if you took it from pot to SAB. Like from as little distance as possible lol.
Hard to do precision work with a butter knife though.
Yeah I’m using a scalpel with disposable blades. 2 to a session. And I’m back to using 2 brand new blades every time. I changed my routine and got contams so overkill is the word again lol.

Yeah that’s what I’m thinking too. I got a big bag of individually sealed syringes. So I definitely get the vibe. It’s just such a shame to have to burn through our materials so quickly. As long as the pot is closed. And it is at an absolutely rolling boil. I use tongues, grab them out of it, and then I just throw it onto the metal rack inside the Sab. Let it cool off inside.

This is why I LOVE my agar punch. Lasts forever. Literally. Sometimes I use my scalpel if I want bigger pieces but a single blade will last me like a month haha

I hear about people using the same blade for like half a year, and when I did it I immediately had problems after maybe 3 SAB sessions.
Now I’m not saying that was exactly the problem. Flame sterilization should have been good enough, but everywhere I cooled my blade, and actually the inoculation loop on one plate, I grew mold. Kinda weird.
So I’m going my back to my wasteful ways.

I'm on my 2nd blade in 5yrs. Just changed it this year cuz the old one was too bent. But I can certainly understand tightening precautions when things aren't working.
