SAB joke

They're all going to laugh at you! One of the reasons why I refuse to use a still air box is that I have terrible shoulder arthritis and torn rotator cuff. My arms just do not articulate well enough through holes. Needless to say it's not very ergonomic for me and doesn't come easy to do SAB work. The best I could do was turn a tub on it's side and wrap the front with glad wrap, leaving a open work zone in the bottom. I know not the best idea but the plates weren't exposed to as much open floating dusty dirty air and I could move my arms a little easier. I wanted to try agar plates again, but I did make some more mistakes this time. I used a whole 11cc LC syringe to 10 plates, way too much liquid per plate. The second thing is that damn parafilm. I can't work with the stuff to save my life, the biggest issue is the getting the backing off. I did see a couple good tips where folks said to rub it between the thumb and for finger to warm it up with friction and it will separate easily. Another feller said to put it on top of a warm light to get it to be more pliable. To be honest I think I'll just cut down glad wrap with a samurai sword to the height of a plate. Seems like that would be easier to work with. Plus my hands are unstable. Ended up with 3 plates out of 10 with some LC lid backsplash which I'm surprised it wasn't more with how much I shake with shoulder grinding and lack of parafilm experience. I left the 10 inoculated plates on a single level unstacked inside a clear latched tote hoping we don't get a bunch of condensation. I'm not going to give up like I have done before, at least three times within the on and off again myco hobby in the course of 2 years. Everytime I do try, even with just cultivating molds and bacteria I get a little more experience and learn something else. I'll get 'er nailed down at some point. Ultimately I really would just like to be able to clone out pieces from desirable fruits on plates and get the trash out before culture or grain introduction.

Not that i would know but if using mss to agar, if you shake the syringe properly to disperse the spores, you only need a tiny drop of solution, even a drop may be considered too much. The hitchhikers guide has excellent agar streak pattern examples which maximize the small amount of solutions potential across the whole plate, a little goes a long way. excess water leads to contam in agar.

Yeah some are flooded, oh well. Trial & error, but a couple of plates I spread out with just a few drops on each, seeing the need for less fluid right away without even been read. Sometimes the ol' conscious is a great guide to what's wrong. I thought these are getting to wet. Thank you for that, I'll look it up.

i like the work around SAB you made, i would try to get a small platform to raise your work area off the bottom of your SAB, the contam settles to the bottom and working down there could stir up the air and contam, maybe a sheet of plastic across the top and bottom with the middle section open?

Excellent start. Glad to see you made a sab. Spent plenty of time at a seated sab. I can't sit for long at a sab, back bitches. I built a (or is it "an"?) Sab that is for a standing person. No pain/discomfort since. Just a thought.

Interesting remark, with the raised kitchen counter island, it's actually opposite for me but I totally see your view. Since it's so high I thought when wrapping the last plate, oh how much easier this would be seated in the bar stools. Seems that finding that perfect working height is paramount. Well no big deal, the specimen was gratis and the plates local, but I definitely see, Mvlted's position point about a drop will do ya. The lids are already sopping wet so I'm probably going to pitch them in a day or 2, and chalk it up to experience. I also appreciate the comment of shit rolls down hill or basically all the dirt settles at the bottom. The middle ya say, well it would be easy to set a working table of some type of fabrication in there and put a condom on the top and bottom..... hmmmm, very very interesting. I'll get another specimen and batch of plates and try a drop each (uh, like 1/4 cc, dumb dumb). Yeah I know pretty sloppy mistake on my part, that could of been avoided. In my defense as I have shared in other places, I learned cannabis (I realize a different animal) by trial and error. I started with a lil bitty grow tent, a compact florescent and ended up with a small ten count of 1000w, climate controlled grow op, organic top shelf, period. Not forgetting to mention the bushes we grew outdoors at grandmas mountain property. Had to put up a perimeter to keep the elk and deer out, apology for ego rambling but it was a nice site to see. Anyways I'm not wanting to cash crop shrooms, it's just my point, I learn best by phaqing up. The end goal is a sustainable tub or two with my own prized clean genes. Heck, we give away shrooms and shroom products around here.
