Some questions after seeing different expressions on plates, from same mushroom
    #28612925 - 01/07/24 11:38 AM (4 months, 7 days ago)

Same mushroom. Same agar (kind of). Same storage place (though stacked upon each other). Different appearances on the agar. Please see the photo beneath.

So, this gave me some questions. Maybe someone can lead me to some more understanding.

1. Does the part of the mushroom where the biopt is taken for the cloning, significantly impacts the appearance/'behavior' on the agar plate and or the genetic expression later in it's life?
2. Does the volume of the biopt has a role?
3. Does any of these ideas make sense: 'the mushroom does what it wants, and phenotypes doesn't mean that much in forecasting. Nor do genetics'.

PS: the plates (actually containers) for sure will not have 100% identical agar+LME+fructose+dye content, as for this batch I filled every container separately with a measuring spoon of the ingredient. So for sure the exact content will differ, which could very well been the root cause for the differences. But still I wonder what others think of my questions and where to find more about them.

PS2: the plate without growth (lower right): there I pushed the biopt too deep in the agar. I think I suffocated it (?).

PS3: the plate on the upper right: there I placed more than 1 biopt.

i could be wrong but i believe the very symmetrical ones are  legit isolates  (monokaryotic) and the others have dikaryotic hyphae

Interesting. Could that (having both mono- and dikaryotic) happen when cloning the same mushroom (like in from the fruit)? (excuse my unknowingness).

I don't believe so. The fruiting body should be made of dikaryotic mycelium or it wouldn't fruit. At least that's my understanding of it. This could actually be contamination that the mycelium is fighting in some of the agar samples. There two with great looking rhizomorphic mycelium that look clean to me. The others appear to be fighting the contamination and that last one looks like it lost the fight to me. But I'm not an expert.
