Any Tips on Agar Transfers
    #28626868 - 01/19/24 08:27 AM (8 days, 23 hours ago)

I've been working with agar for about a year now and for the most part I've had plenty of success and most of it seems fairly straightforward. But there are still some things I'm not sure about.

The above pic is after the first transfer from spores. Growth has been tomentose throughout which doesn't bother me, but if I go for another transfer, is there anywhere in particular someone more experienced than me would go for? The thicker growth or the more uniform bits? Are there any obvious signs I'm missing here?

First,  what is cg ? Cambodian Gold?..

Is it cube?

What's is agar recipe?

Still looks A little small to be xfer'n for isolation sake.

what is your intentions ?

Such as finding the best iso for big fatties/fast/ canopy pinsets/ potency/ special phenos....

The reason I ask , is unless that transfer is dirty or needs cleaned up, consider;
that it may be beneficial to have such a diverse gene pool from MS.

One could take a few different sectors and send them all to their own SB in order to find the clones worth taking to the next step or spore prints.

But generally speaking, the best myc to look for is the fastest/aggressive..
HOWEVER, not always does the first good looking Rhizo become the best culture for any particular area/environment.

There's a YT of time lapse video on agar selection/sectoring and the winner was definitely not among the first choice picks at the beginning.

Also if looking for more rhizo not that tomentos is bad....
A low nute agar may help.

Also with colored agar look for the places that are becoming lighter in color, as the myc feeds off the nutrients the coloring get ate up as well.

if we have any pending trades or you never received anything that you were expecting send a PM with details.  I've had a lot going on, and may have overlooked something as well as USPS snafu's.

Yes, Cambodian Gold. cube. Recipe is 1:1 agar/LME and a small amount of nutritional yeast. Green food coloring added.

Okay, I hadn't considered that approach. Definitely makes a lot of sense. I more or less transfer to hedge my bets in case there's some contam I just don't see. Honestly, I don't know if I've ever seen contam on a plate other than the odd pour where something snuck in.

That's where I ended up taking a transfer from. The day after I originally posted this that section looked much better than the rest.
