Regrouping My Mycology Process 2

Update... Had to make some adjustments to my process here...I have way too much contamination going on. I also think my impatience isn't helping anything.
I have a couple other posts going on my progress as a rookie mycologist... How does this look getting ready for -FRUITING- Pictures Of Colonizing Grains Long Grain Parboild Rice Photos through jars POPCORN
SO I REGROUPED.. I went through and removed all contaminated jars also a good scrub and cleaning of my area. THen...
I am a rookie at this and of course did a lot of reading ahead. Watched the youtube videos and whatnot. I got my hands on a spore syringe and it was on. Now in hindsight I notice patients is a must! Cleanliness against contamination...a must! Trial by error, a must!
Fast forward...I have some LC cultures I was able to make from B+ spore syringe vendor. I also, tried inoculating spawn brown rice jars and some Uncle bens Instant rice paks. Hindsight, I was trying to inoculate my jars using the oven tek method...bad decision as I used this method in both of my inoculation sessions. From my results probably a bad choice! So I pulled me SAB together for future sessions.
I have also worked on my tek for preparing my rice spawn grains, which will still need some experiance and work at, but I am getting better. Also working on different LC culture recipes. I managed to get a good process for AGAR to start to use in my processes. I was suggested so I decided I needed to jump into getting used to using it. Found a kewl Potato Dextros Agar recipe. Which I was excited worked well on my first attempt at making it! Hurrah! Pictures of my successful AGAR dishes

Was kinda tricky pouring in my SAB but it worked!
Now...I can pull from my original LC from my vendor spore syringe...and actually see what I have here...
What methods do you guys use to get a LC onto AGAR??

Agitate the culture and suck some up into a clean syringe. Put a drop on the dish.

Thanks, I was thinking that was enough. I appreciate the reply. I have some LC to get onto AGAR

Currently I wash/rinse rice, add rice to already boiling pot of water, boil for around 17-20mins. Drain and rinse with cold water, place on flat surface to air dry for about 1-2hrs. Jar it up and PC for 120mins, cool in pot for 30mins and then remove.
How do you do yours?

I don't use rice beyond grinding for flour. It's just always seemed like it would be really easy to have the grains become too mushy. I'm kind of amazed you're able to boil it for so long.

Quite similar to grain prep. Very nice
Someday I wanna try wild rice. I think it would look cool.
As for transferring LC to agar, yeah use a clean, empty, new spore syringe (blunt-tip syringe). Or you can also use a new long glass eye dropper or glass pippette.

I am actually thinking of switching to whole oat grains...I can get a 50# bag at Tractor Supply for like 27$ and that would last me quite a while. We will see. If you cook it correctly, the rice actually doesn't turn out mushy when done correctly. But I want to make my way through all the grains until I find one that works well for me

THanks for the tip Yeah its pretty simple..tried soaking it overnight for another bacth of grains, tonight, the only difference..we will see how those do! I agree on the wild rice thing..it would be cool to watch colonize

Seems as though my new LC syringe from Myyco, shout out to them, is going great! Great product, WOW! I enjoyed getting it! Anyways, great all the way around!Very Pleased!

Was able to make a LC Jar and some others. Seems to be vigorous! Just after three days.

I even through some to AGAR to see how it grows out all for educational purposes;);)

I am not happy with my B+ spore syringe that I started all of this journey with at first....I am working on narrowing this one down with AGAR. I have a few plates that I am trying to get to LC Jar that I can spawn with. Sad when the first strain you wanted to start with isn't going as planned..maybe it was contam....??
Anyway I tossed all but one LC Jar and One popcorn jar that looks to have some viable mycelium I can throw to AGAR and narrow a good culprit. Fingers crossed.
I didnt start off with AGAR, but I am glad I have started the AGAR journey

Update... Seems as though everything is paying off in a more defined and clean environment! I am seeing some awesome growth on my Koh Samui Super Strain (KSSS)! I did find some contamination that I need to pin down was it due to the process or extra moisture.
Started a fresh Jar of B+ LC and NOC'd with some good growth in my SPORE to LC jar, so far the growth is clean and promising!
Use of the SAB definitely helped my process and mitigate contamination! Some picture...

Some good growth and some contamination on three jars unfortunately....but I will note I changed my dry time on the grains before loading up on the second batch of jars and those seem to be doing well!

You can't remove contamination from jars. Peroxide and surgery are options for bulk substrates and it's not even guaranteed for those.
Better to make new jars and try again.

I agree. I think it was a moisture problem on these three jars...Everything I Noc'd up this last time has zero c'tam.

Hey everyone! Just wanted to throw an update your way!
I have been busy. Have some success with my mycology journey!
Most recently, I have been able to move my spawn to bulk

I also wanted to get some experiance with grain to grain transfers so I have 6 jars of Popcorn ready to be noc'd up!

I am hoping that the growth is quick. So far I have seen, spore growth, mycelium for liquid culture growth; so hoping that the grain to grain goes well!

Yes but for the record most people put tubs into "fruiting conditions" at the time of spawn.
Personaly i would take a razor and trim that liner all aroud within an inch of the sub but if you don't think you can do it without molesting the sub too much then just leave it and add your fresh air.

I did think about just trowing it straight into fruiting right after mixing spawn with the substrate however, I wanted to see how it went. I do need to trim it down better:)
Tonight after work I am going to throw this one into fruiting....I did get a three more tubs going last night...now I didn't throw these right into fruiting either....
Is there any real benefit from throwing them straight to fruiting instead of letting it colonize the substrate??

It's more like there doesn't seem to be a benefit to suffocating them during colonization.

Here is some pictures of my hard work at work finally paying off!
