Just followed this tek. The centrifuge tubes work awesome, lots off room. Question, does the wood need to be exposed a lot? When I tipped mine, had to run errands, got back and just had a tiny bit of wood poking out of the top of agar. Should I redo with more wood exposed?

I have seen multiple people say to put slants (or petri dishes if you use those for storage) in ziploc bags and then in an insulated lunch box. What is the use of putting them in ziploc bags?

Works similar to parafilm or cling wrap. Provides ge while keeping contam out and keeping the plate together.

But we already wrap the top of the tube with parafilm or cling wrap..

Does anyone else find that some of the slants get a really tiny bit of agar gunk in the lid after PCing them? Is it likely to cause any problems? I'm thinking that the mycelium is never likely to colonise it, and it might start feeding a stray contaminant at some point! Any trick to avoiding it? I am currently sterilising a couple of replacement lids for two of my slants as it makes me nervous.

I haven't found anywhere else that recommends the wood, not saying I disagree. I'd like to know more though!
The other often-referenced recipe I see is BAM Media M85: Long-term Preservation Medium but the idea of 30 grams sodium chloride seems really sus.

Slants aren't even necessary, you can just add a wedge to sterile distilled water in a centrifuge tube, or some other appropriately sized vessel. Dehydrated grain is also a cool method.
Plates can last over a year in the fridge, and LC broth will keep your culture alive in the fridge for just as long, if not longer.
Slants seem outdated, imo, but still work.

what's longevity of wedge in distilled H2O @ appx. 45 °F? To use the genetics, do you just put the wedge on agar?
