Too Many Mushrooms!
(I Hope)


I decided to play catch up after several years away from the hobby and found five new species to work with. I'll post a weekly update on each until I get fruits (and sclerotia) from each of them. I expect the woodloving mushrooms will take about a year to grow outdoors, so updates will slow down during their dormant season. I'll also be trying a (likely to fail) experiment with P. Allenii this fall as well. The first update will come tomorrow after I've had a chance to do some SAB work and take pictures. With each update I'll try to include dates, pictures, upcoming plans, and post links to any Teks I'll be using.

I've worked with all the required substrates(manure, straw, coir, and woodchips) while growing cubes and various types of oysters- but each of the species below are brand new to me.

The Species:
-P. Cambodginiensis
-Ps. Natalensis
-Ps. Tampanensis
-Ps. Allenii
-Ps. Azurescens

This is what the plates looked like on the morning of 9/5. Everything looks remarkably clean, so I'll be taking transfers and making LCs of each (except the Ps. Azurescens taking it's sweet time).
I'm a bit worried that the Ps. Allenii might have some hidden contams riding along, but worst case scenario I should have clean plates I can return to after this transfer.

Update 1

I'm going to consider 8/31 day 0 for this log since that's when most of the dishes were inoculated. The only outlier is Ps. Allenii which along with Ps. Azurescens is really in its own category time-wise.

Days 0-7


Day -4
Ps. Allenii arrives on it's own and a petri dish is inoculated.

Day 0
All other species arrive and spores are placed on dishes the same day.

    -P. Cambodginiensis
    -Day 3 Faint growth
    -Day 5 Solid growth slow but steady.

    -Ps. Natalensis
    -Day 2 Faint growth
    -Day 5 Aggressive growth. Very dense and rhizomorphic from the start,
      lots of aerial mycelium as well as what's growing against the plate.

    -Ps. Tampanensis
    -Day 2 Fast but somewhat wispy mycelium.
    -Day 5 Slowed down for a day or two but is back up to speed now. Less 
      dense than Ps. Natalensis, but probably the fastest growing of the
      bunch.

    -Ps. Allenii
    -Day 4 Faint growth
    -Day 8 Growth is dense and steady but not especially fast or
      aggressive.

    -Ps. Azurescens
    *Crickets*

Next Week
Transfers of the following were taken to agar and LC. LCs are being stored at 75F~ and stirred twice a day using the magnetic stir bar. Hoping to have usable LC in about a week.
-P. Cambodginiensis
-Ps. Natalensis
-Ps. Tampanensis
-Ps. Allenii

I meant to take pictures before I took transfers, but that didn't happen. Here's what the plates looked like after I took my wedges and a picture of the new baby plates(sorry for the picture quality, I'm looking for a light table that should help with that quite a bit.)

Plans for next week are to hopefully inoculate some jars! I should test my LC's on agar first, but I'll probably just inoculate a few jars and petri dishes all at once. I don't mind chucking a few grain jars if it comes down to it.




Methods

I'm going to save this methods section after the updates each week. There is nothing special about these methods and I don't recommend anyone copying them. However, if one or more of these grows fails I'll have a good record of what might have gone wrong.

Grain Water Prep
Both my petri dishes & LCs are made using left over grain soak water in place of malt extract. Normally using grain water results in some funky looking LC(hazy with lots of sediment.) It works just fine, but these steps make for a much more transparent LC/Petri dish.

1.PC Grain water for 15 minutes. PSI is not important and no 10 minute purge.
2.Allow the grain water to sit in a large measuring glass or pitcher for about an hour
3.Pour grain water through a coffee filter being careful not to let any sediment leave the measuring cup.
4.Use right away or freeze in an ice cube tray and store forever.

Liquid Culture
-50% processed grain water
-50% water
-1 Magnetic Stir Bar(optional)

1.Combine all ingredients in a canning jar with a suitable lid(picture below)
2.PC at 10 PSI for 35minutes
3.Inoculate within 2 weeks


Agar
This recipe should make about 30 60mm petri dishes
-125ml processed grain water
-125ml water
-5g agar
-1 magnetic stir bar

1.Funnel agar into a 375ml Bombay Sapphire bottle with a hole drilled in the lid and a filter disk sticker over the top.
2.Add magnetic stir bar
3.Add 125ml very hot water and stir on high speed until agar is dissolved
4.Add 125ml of hot, processed grain water
5.Screw the cap on, cover with foil, and PC at 10PSI for 35 minutes
6.Cool to 135F and pour plates inside SAB
7.Once cooled to room temp, inoculate and bag petri dishes individually or store several unused plates per Ziplock bag at room temperature for weeks~

Update 2

Days 9-15

Day 13
-The Natalensis plate either has some contamination OR it's just another piece of Natalensis mycelium that hit the plate when I was trying to get the agar off my scalpel. Either way I decided to transfer to a new plate. All other plates look contam free. The Natalensis mycelium grew right over whatever the contamination was, circled in red.



Day 15
It's been about 2 weeks and the Azurescens plate has not germinated. I have a couple backups, some at about 65F and some at 75F, but it's the same story. I'll be looking into germination tips for this species.



Day 16
Here are all the plates 1 week after I took transfers from the initial plates with spores. The exception being the Natalensis plate which I had to transfer on 9/13. They're looking pretty good, so I should have some clean plates to return to if any of the LC's don't work out.

Next Week

The liquid cultures are all moving along at varying rates. Unfortunately I did not use the same recipe to make all 4 liquid cultures, as a result only the P. Cambodginiensis is clear enough for quality photos. The only difference between the two recipes is the extra pressure cooking step mentioned in the methods section of update 1. On the bright side, it took to the LC the fastest which really surprised me considering how fast the Natalensis and Tampanensis were on agar. Part of me wonders if the extra pressure cooking helped break down some of the starches in the grain water making it easier for the mycelium to digest- but that's just speculation.

P. Cambodginiensis LC


I'm testing all 4 LCs on agar just to see if  everything is clean so far. I also made 3 grain jars each of the P. Cambodginiensis & the Ps. Tampanensis. The Natalensis could probably be used today, but a few more days would be better. However, the Ps. Allenii is probably going to need at least another week.


Methods


Grains
Right now I'm using rye and I don't bother boiling the grains. Grains are rinsed in a strainer then added to a pot where they are rinsed one or two more times before being covered with hot tap water and allowed to soak for 24 hours. They are then strained again and loaded into jars still moist on the surface(no fanning or anything)

Grains come out between 52-53% moisture content, dry on the surface, no clumping, and the grains sperate easily once colonized. This also allows me to use a HEALTHY dose of LC which I always seem to have more than enough of after I make it(the jars just pile up...)

Determining Moisture Content
Here's how I figured out the moisture content for my grains, I don't do this for every batch, but I wanted to point out how I came to that 52-53% moisture content that I mentioned above. I'm sure there are better methods, again this is just what I did.

1. Grain in placed on a baking sheet and placed in an oven for 6 hours at 225F.
2. Check the weight of your grain at hour 5 and place it back in the oven.
3. Reweigh the grain at the six hour mark, it should be the same as it was at the 5 hour mark, but if it's lower place the grains back in the oven for another hour and reweigh.
    *For me this meant that 4 cups of grain dried from about 750g to 675g
4. Allow the grains to soak for 24 hours, completely undoing all your hard work.
5. Strain them well and reweigh. Use the formula below to determine moisture content. I'll use my numbers as an example.

Formula for water weight:    Total weight - dry weight= Water weight
Formula for moisture content: Water Weight / total weight= Moisture content

OR

Water weight: 1425 - 675 = 750
Moisture Content: 750 / 1425 = 52.6
