Liquid culture problems
    #2845449 - 07/01/04 12:32 AM (12 years, 5 months ago)

So I have a couple of liquid culture jars and the other day I tried to use one(inject a jar), and saw no growth the next morning

So I injected another jar, still nothing, then the original jar again, NOTHING

I didnt put glass or marbels in the bottom of my jar, could this be the problem???( the mycelium is to thick???)

I wouldnt think that woudl cause a problem , especially because when I shake the jar really hard the mycelium breaks up anyway

None of the jars contamed they just havent grown?????

the first one was started on the 23th

I have used 2 jars(of liquid culture) B+ and lipa, and neither started, what am I doing wrong???? (I have NO CONTAMS, I dont get it! )

Hmmm it has been incubation kinda long, I really was thinking about sticking it in the fridge , but its still growing , so I assume its not dead.

I am assuming you saw growth in the liquid cultures. Any visible amount of cells should spawn growth -- if you left the liquid cultures sitting for more than a week after they were colonized the cells could have autolysed and died, but more likely is that the jars were prepared in a way that would slow growth (i.e. too thick or wet of a substrate) and/or that you simply haven't given them enough time to show colonization, especially if there was a small amount of inoculum or you injected it into the center of the cakes, where the growth would be from the inside out. Keep in mind it could take up to 2 weeks (or more but not in most cases) to notice growth. If it's simply taking to long you just need to promote faster growth, i.e. more cells inoculated or better substrate formulae or better growth conditions.

Given these factors, the only other possibilities lie within the conditions of growth for the jars (if they got burnt, etc.) Contamination should either be visible or have a smell.

" if you left the liquid cultures sitting for more than a week after they were colonized the cells could have autolysed and died"

the jars seem fine, and the mycelium grew  well over a week, that beig said, I better get my newest jar(cyan culture) in the fridge soon

Thanks Micro, awsome info as always.
