I stored them in cool airtight dark place. Is not it. They are very much potent but not as original. There wasnt any contamination on them nor in the box till the very end

Could be a storage issue. Light degrades potency. Freezing can also degrade the actives. Also another factor is contamination, I’ve long suspected that bacterial contamination can override certain expressions, certainly pigment can be one. I suspect potency can be another.

This is the thought provoking content that keeps me coming back. Bacterial contamination is interesting food for thought.

I think you've misconstrued my post a bit. When I say bacteria can effect things I'm not talking about massive pooling blooms. I'm talking about that tiny latent bacteria that can lurk in the spawn almost invisible to many people. In many cases that bacteria can be a vector for mold later but in the cases where mold doesn't get a foothold the bacteria is still there causing issues. I've had isolated reverts occur in leucistic grows where only a couple fruits reverted. This indicates to me that small amounts of bacteria can effect outcomes even if the fruits are otherwise normal looking. Certainly in my case the grows where that happened were with clones which means the traits weren't being consistently expressed.

If you have spotty potency from clones that should be providing stability, that would be the first place I would start looking. Increasing PC times for spawn by small increments won't be a major undertaking but could prove to be the difference maker. Just my hunch anyways

Also, stating that you've cultured, grown, dried, stored, all perfectly executed yet still cannot troubleshoot tells me there's a flaw in your perfectly executed approach.

Potency is genetic, therefore your issue is the expression of the genes responsible for potency. Either you cloned a weak fruit, or as Pasty pointed out above, incomplete sterilization.

Go back to the basics. Test your clone, ensure you have pure culture, then maintain with aseptic technique. Increase vent time and sterilization cycle for good measure.

With all of thhe info you've received, you should be able to fix any issue with your potency issue. Take into consideration your own nervous system with regards to repeated exposure leading to tolerance.

Here's a thought experiment:

You take 3.5g, this dose consists of 3 different dried fruits, all consumed simultaneously but one of them has been cloned.

The result may be a powerful trip leading you to believe all fruits are equally potent.

Every clone must be tested individually. Take into consideration that your perception of potency will change day to day, your nervous system isn't designed with the sensitivity required to test miniscule variations in  chemical concentrations.

Great point on tolerance stipe, I myself developed a massive tolerance over time even with waiting a week or longer between doses. It's my belief that to truly get to baseline is closer to two weeks or more depending on the individual.

I dont agree with you. Bacteria prevents mycellium from growing/fruiting. It doesnt affect potency.  I think i must dry them at 40 Celzius max.

I don't know, man, you might ruin the batch running that hot, I wouldn't go above 10°C. :trollhide:

Potency as experienced by your meat machine is subjective, you're not a mass spectrometer. "Very much potent but not quite" sounds like you're chasing sensory perception.

If your source materials are improperly dried fruits, stop being a noob and get better source materials. Cultures are ubiquitous these days, ask the guy to hook you up and maybe learn how to use a dehydrator properly.
