Silly Experiment - Attempt to Make "Cleaner" Prints * 5

So, we typically do our best to make our prints as clean as possible, but nothing changes the fact that, short of plate and jar pins, we're growing those mushrooms and their spores in open air.  Nevertheless, I make my prints and swabs in my SAB, wear gloves and a mask, follow clean technique, etc. 

Feel free to call me an idiot or chime in and say its been tried, but I cut the very top off of a couple fesh caps in an attempt to reach "deeper into the gills" to portions that are exposed to perhaps less airflow, contaminants, etc.  I'm now trying to see if these trimmed down caps will drop a print. 






Not performing any of this in a sab or with sterile blades, first just wanted to see if I'll get any spores.  Doing this on paper as well, as I figured it might be a bit easier to see.  Also, am going to see if one of the lower halves with exposed gill segments will print upside down.  Placed them in a little shoebox...



Initially, this has made me think it's way too much work to get a print.  Precise cutting, potential to just add more contaminants, etc.  BUT it has made me think this could be a good technique to get cleaner(?) swabs.  There would be less precision needed.  Basically, cut away from the top until you see gills, and swab away.  Of course, then you're also dealing with the fact you've cut through the outside of the mushroom, as well, negating any flame sterilization of your blade, etc.

Random thoughts, but figured I'd share.  Even though it may be extra work, I can see utility in gathering slightly cleaner spores in cases where one might be going through many generations in an attempt to stabilize a varietal; it might save a little time in the long run.  In a cross of a couple strains I'm working on, I have several times put spores straight in sterile water off a print and sent it to grain, while also cloning that mushroom, cleaning it on agar and doing it properly.  Recognizing that's not the right way to do things, but also trying to get through generations quickly enough, it's worth a gamble of a single jar to me.  And it works sometimes. (required caveat do not go direct to grain with spores!

Anyway, I'll post if I get anything on the paper.  Perhaps it's a cleaner print.

Alright, so roughly 16 hours later we do in fact have spore prints.  All three left spores on the paper, as seen below.







Are they light?  Sure, but its more than enough to play with.  Would these be any cleaner than a typical spore print?  I dunno.  Could set up some sort of control, even exposing whole mushrooms to contaminants, etc and test on agar.  Not sure I'll have time for that in the immediate future with current schedule, but we'll see.
