Cleaning water agar plate from contams
    #28391533 - 07/10/23 04:55 PM (1 year, 6 months ago)

I recently took some clones from pan. cyan into water agar. The clones are filling the dishes slowly. Some of them have visible bacterial growth some of them have slowly advancing bacteria with mostly clean mycel. Some even seem to have no contams at all. So the next step would be to transfer them to nutrient agar right? I already prepared an oat flake juice agar.

The mycelium on the plates is wispy and thin.

so here's an obvious contam am I correct?

And here hopefully visible with mycelium and contam in center. I know it looks like there's contam in the upper part of the pic. But thats just light glare from fluorescent tubes.

So this is my first time cleaning agar. So I can observe that the slimy part in every of the contaminated plates starts in the piece that i cut from the shroom. So starting from the same point as mycelium. But advancing much slower. (both grow really slow, but that must be because of water agar) So I should just avoid the slime when doing the transfers?

Yes you definitely want to avoid the slime which is bacteria. Let it grow out so you can comfortably take a small piece from the leading edge of the mycelium while staying well away from the contamination and transfer to a nutrient agar plate. After this it could be already clean but you might want to expand the clean mycelium to a few more plates to have some for inoculation and some to keep as backup.
