Depending on the grain, hydrating would still be needed. Smaller amounts of millet and such should hydrate and sterilize in cycle without the prep but larger grains like rye, etc would do better prepped to ensure theyβre fully hydrated and sterilized in cycle. If no grain was used a nutritional component would still be needed for the readily accessible carbon and nitrogen.

Grain can be shaken which speeds things way up.
Those bags still have grain prep, theyβre just hydrated and then PCd along with sub instead of on their own.

I'm here to tell y'all you can shake AIO containers just as well as straight grain. Quote:

Also, you can still shake all in one grows. Haven't seen much of it on here but on reddit I always see people say to let the grain colonize before mixing with the sub in all in one bags but the bottle grows I do are mixed from the get go and I shake them bitches like a baby that won't stop crying.

Thanks, I think I'll run some Mudafuka bottles again for the sake of it.
SirPsycho , is there any reason to use bags instead of PP5 containers? Or just stick with PP5's?
Also, I keep growing these weak ass looking plates - are these contaminated?

Yeah, plenty of reasons. Bags hold more stuff, you can fit more in a pressure cooker with bags cause no gaps, no need for a fruiting container or topper for the jar, less clean up, easier mixing.

Can a TC weigh in on this thread: https://www.shroomery.org/forums/showflat.php/Number/27351106
It was started by a TC and another TC replied in there so I figured I might as well keep the TC train going.
I'm in a similar situation as Hindsight, loose unmodified lids, bone dry grain, slow colonization (8 days post inoc, 4 days past shake, 10-20% colonization right now), RH of 45% and a pretty warm grow room.
I think I'll take a couple jars out of the batch and inject them with 10cc of sterile water like Hindsight did.
Just wondering if there's anything else I should know regarding this. (Dry grain/wet grain, loose lids, tight lids, GE etc)

i lost a entire collection of spores to travel from one city to another, spores were in moving truck with other mycology stuff 1600 miles don't know what happened nothing germinates from any of spore print and neither from swabs

i am not going to refrigerate cultures either.
my question is about having a backup of spores for long time, in case i have to move this wont happen again.
i read best way to save spores for long period of time is to put them in distilled water. now claims are spore remain alive for decade is this correct?
also how to get spores from variety of shrooms that don't drop spores? like whats the protocol for ghost and P.E to save spore for long time?

My experience seems to be that lifespan of spores is random based on culture. Keeping spores at room temp in a dark drawer most work fine years later but occasionally Iβll get some that are completely dead after like 5 months.

i need to secure some good cultures then, and storage is the same as you said room temperature inside a drawer its a dark place.
i was skeptical about the claim spore remain alive for decade in distilled water, if its weak genetics they will be dead eventually in couple of months.
thank you A.K.A

Has anybody seen this Dr Myc's, any idea of the ingredients or what CFU's refer to exactly. Is this just a gimmick or has anybody used and had good reviews?

Yeah thereβs this wave of people using βnutritional subβ now on discord and Facebook. I donβt understand the appeal of basically creating sub par manure using coir and nutrients and paying extra to do so.
Some people just get sucked in to that stuff. I know one guy who had great grows using coir and wbs and then started buying βnute subβ bags online, had no measurable increase in yield, but continues to buy it. He also uses over 20 bottles of nutrients for weed.

people tryna bring plant knowledge to fungi.
with weed ive learned dirt is king. its almost all about the soil and of course light. shrooms just dont give a fuck.
people wanna over complicate and instead of taking credit for pushing out good grows they wedge a useless product or theory between themselves and legit praise.
its weird as fuck to me.

Fellas, give it to me straight. Am I dealing with enmeshed mold here or am seeing things.
I've been trying to get something out of these 4 prints for two months now and I got diddly squat. Either I get bacterial jars or I get mold before first flush. Unless you count that one shoebox that decided to literally cover every square mm with pins and aborts. Still doing it, just pumping out pins and aborting them.
Anyway, am about to just throw in the towel and trash everything. I'm just fucking spent dealing with this.
Here's a selection of pics, I got tubs full of plates and they are more or less the same.
New T1 from a new germ plate run I did on the left. Yeah I know, bubbles, I fucked up my pour. Plate on the right is an older gen, that looked like the one on the left until it matured and had a fucking chestburster event with mold exploding out of it. Scope view of the thick ass hyphae at 7.

Another new T1 plate, another print/variety.

Will probably end up looking like this older gen.
Or this
Or this
Or eventually also have mold burst out of it.
Or look at this guy, I think that's a T3
That one actually had a nice thick spiky silvery hyphae shoot out for the lid, but it got flattened for some reason when I put a cup of hot water. Here a scope pic

Anyway, whatever more pics:

T1 from a fresh germ plate And it has that massive halo of extremely translucent myc, only visible with incident light that it reflects back.
Whatever cube myc growth is there, it's covered by this fucky barbed wire looking myc (sorry shit scope pics, phone just refused to focus on eyepiece. Just imaging long straight cube myc in the back, jumbled mess of whatever myc on top)

Even clean rhizo will eventually get engulfed by this weird aerial shit. Maybe it's normal but it looks like a jumbled mess on the scope and it makes this little clumps as it gets older.
That shit at 9 is not light reflecting, it's just aerial myc consolidating or something. Makes little clumps. The bottom right area looks uncolonized but with the scope you can see tiny tiny little hyphae sticking out here and there.
And ofcourse this type of fuckness. Yellow stuff is just hotbreak boogers from the LME.
Plate after plate after plate, just starts off scalloped, stalls and then random patches of myc popping up here and there. Never sporulates though and never starts as satellite contam. Always with the transfer. Never seen bacterial biofilm either.
What do I do? I tried a 1ul inoc loop and a cell spreader to separate the spores, still not seeing 100% clean stuff.
Josex pokes/brf pucks? But I'm not seeing any bacteria.
