spawn bag check??

OK so i made a bag of 30 bags. I use the unicorn 10t with millet. I used to use a no soak method of 1kg millet to .5 kg H2O. I had a ton of them turn green. SO i tried a 24 hours soak with 2.5 hr PC at 15 psi. The reason i cook so long is that the bags are 1.5 kg of stuff. They are around 5x5x4 when cooled, so i need to cook them long enough to kill everything. SO this batch i did 20 bags.
How should i go about testing a bag or 2 for contam? Just seal it and let it be? Or should i just let it stay in that "vacuum-packed" way they come out?
I have been having a ton of issues with trich getting into everything. Also if anyone knows a good LC tek.....i have looked through a bunch.....but nothing very detailed.....and nothing that covers using stir tables.

Got any pictures of agar, grain & bags? We could use more info on when/how things are contaminating to dive in deeper into the root issue.
LC is the biggest contam risk that I see in your process, so you might want to take a step back from it and just do agar > master jars > bags. You can abuse the hell out of clean spawn and still get fruits, but spawn weakened by bacteria/environmental stress/other fungii can die off quickly .
If you're worried about the grain sterilization, seal up a bag and let it sit so you'll have a control to check against but sterilization failures are usually pretty obvious by how fast the entire container will contaminate from everywhere and not only growing from the colonization points. FWIW I do a little over 3 hours @ 17+psi for 4lb wheat bags and have never had a bag fail due to sterilization issues. I wouldn't rely on the bag vaccuum for more than a day or two as they tend to develop leaks if handled.

so i really need step by step for using stir table lc's. For some reason i think i let it go to long and it gets cloudy. tons of mycelium at bottom. but just cant risk it. also, anyone have any guides to spot comtam in bags? No the obvious stuff. Like the different type of white growth. In the jars i could easily see it. Instead of looking like cotton, it looks like marshmello.

Those lcs are dirty. Use .2% LME and you’ll get a clear broth that is easy to inspect.

also, for millet, is the boil step required if you are 24 hr soaking?

i had those lcs going for 14 days already. They were a perfectly clear, slight yellow. Yeah i used a .2 lme but i used .5 g/500 ml. soy peptide, so maybe the extra nuts gave the bacteria something to get it on?
Super interesting......the one on right, had 10 mg gentamycin in a experiment to see if i could forgo contam. mycelium developed with in 4 days, nice mat of 5 mycelium by day 5....and perfectly clear. like perfect. but then i just let them keep running. like 2 days ago, the tops of those LC's was clear. The mag stir plates make every happen very fast and sometimes i think they accelerate the process too fast.

Sorry, I should have said they look dirty. The stir plate should have no impact on contamination. I’d recommend just using the LME only. All that said, sounds like you also need to shore up your bag prep. Instead of making such large volumes of grain and lc, maybe get the basics down, like some quart jars and agar inoculation.

As mentioned, you should increase your pc time if you’re cramming bags. Sealing a couple up and letting them sit might be a good plan to rule out whether or not the issue is the grain. But like i said, maybe work smaller to ensure you have success with small volumes before losing a ton of bags. That LC doesn’t look good to me, so that could be the issue or perhaps something is off with your technique in inoculating them. Is your syringe sterile? Are you flaming the needle before aspirating, are you working in front of a hood? All that stuff. Something is obviously going wrong and if you can’t make clean LC and noc some jars with success, maybe you shouldn’t be going huge until you’ve mastered those things on a smaller scale.

fair, but how long should LC go anyways? Bc honestly, 2 days ago they looked clear. I am using a hood, flame sterilize every time and each i use a fresh needle to avoid the cross comtam.
Just trying to understand why it stays perfectly clear, but then clouds.

What is your LC inoculant? Your technique can be great but if you are adding cooties to the jars, nothing else matters.

Honestly I think the problem is in the plates. They don't look clean at all. There is more than likely mold in your plates and everything else is screwed from there.
