Is my liquid culture doing ok

I am new to the hobby and wondering if my liquid culture is doing ok

If you're new then my guess would be no. Those large surface bubbles aren't a good sign. I'd recommend not using LC at all until you've learned the basics.

The rule of thumb should be: if you have to ask if it's ok, it probably isn't.

If you insist upon using LC, this is how I do it:

Give that a read.

Test it on agar.

The bubles are often an indication of microbial activity in the broth, with the exception of stirring/agitating.

I’m not worried about the bubble like I said pic was taken right after agitating the jar but I have brown streaks it’s penis envy if that helps

I don’t have any agar was considering just buying more syringes and using that to inoculate my grain spawn every liquid culture I do I end up with brown cetament on the bottom I inoculated another one sat looks better butt I’m hoping no cetement happens idk what I’m doing wrong?

What youre doing wrong is that youre attempting to do LC without first mastering Agar. All LC should be tested on Agar, LC is not a 2D Medium that you can see contamination in.

Spore syringes to LC is frowned upon due to the likelihood of contamination.  You haven’t posted the LC recipe but the sediment is likely ingredients but could be something funky…your pic leaves a lot to the imagination as it’s not very illustrative of what you’re referring to.  Spore syringes to grain are also often a gamble for the same reasons.  If you’re really wanting to grow with grain, start agar work and inoculate your grain and LCs with clean cultures. It can be done cheaply and is not difficult.  Otherwise, you should look into pf tek.  Ziran has a nice write up.

I don’t trust anything I didn’t make myself.  You’re also paying a premium on premade products and will save tons prepping yourself.  Homemade agar media, spawn and LC costs Pennies.

It is a hobby that is founded on the science of microbiology, but don't let that dissuade you. It can be as easy or as complex as you'd like it to be.

By choosing to use LC as a new grower, without agar, you're definitely choosing the hard path.

Start with agar or pf tek depending on if you have a pressure cooker or not.

So what's the established way to test LC? I'm a silly paranoid bitch and I worry I'm going to get a false negative while contaminating the LC.

If I sterilize a syringe/needle in the PC, it gets vacuum locked so I can't really trust it unless I break the vacuum while the needle is submerged in the LC. Is that how you guys do it?

How do you test it then? Just free pour on the petri? I'm talking about home made LC in media bottles, not that newfangled LC syringes that you can buy.:oldman:

I’ve never had an issue with vacuum on a sterilized syringe.  If it’s an issue, order some sterile, individually sealed pipettes.  You could also use a loop.  And to be honest, I don’t test my LC.

If your sterile technique is on point and you understand when you make mistakes, you have a lot more room to be confident about these things haha.

This is the reason why I make low nute, clear LC. It colonizes the broth a bit slower but saves time in the end by not requiring testing on agar.

If the mycelium is white as fresh snow and thw broth is bubble free/crystal clear, without even a hint of cloudiness. It's most assuredly clean.

LC is single use media, imo, so testing is an unnecessary risk/vector for contamination.

This took 7 days:

Each bottle is enough for 10 jars.

:takingnotes:

I always wondered this too, the fact that the process of testing LC can actually contaminate the LC. Always thought that the "test your LC" Advice was logically associated with Vendor LC. And that if youre at a stage in your cultivation where you feel confident in making your own LC (which also means your sterile technique is pretty much immaculate), then you probably dont need to test it.

Question though, when you use LC do you just pour? The syringe seems like another vector for contamination, so pouring seems more common sense to me

Yep, I pour its straight from the media bottle directly into jars or bags. As seen here, disregard the amount being poured, I needed to use it all up

Notice how I keep the foil caps on the lids that are behind the jars receiving first air? This maintains sterility while in turbulent flow.

Oh look, I'm just being a retard again. Just use a pipette tip or inoc loop. Right.

But what if the loop was not properly sterilized at the factory? Or what if I contam it while taking it out of the package?

LC is not for paranoid bitches lol.

I don't test my LC either. :fuckthepolice:

But I also grow mold as my day job so YMMV.

Yep, I pour its straight from the media bottle directly into jars or bags. As seen here, disregard the amount being poured, I needed to use it all up

Notice how I keep the foil caps on the lids that are behind the jars receiving first air? This maintains sterility while in turbulent flow.

:takingnotes: ill keep this all in mind when i graduate to LC and a Laminar flow hood in the distant future ;___;

LC is single use media, imo, so testing is an unnecessary risk/vector for contamination.

:emotionalmoment:

Fucking finally, a TC confirms I'm not insane. You can't test LC without having an  observer effect. It's not as bad as quantum entangled photons, but still you are fucking up the thing you are trying to test.

Just shove that bitch in there red hot? Sure why not. I have some spent LC, I've been thinking putting that shit to agar to see if it was contamed to begin with or if my unmodified jars were the vector. Will do.:boobsmell:
