Reviving eight years old cultures

I'm moving this to a separate thread from the 'The Gourmet Cultivation Discussion Thread'
The posts I'm moving:

Walter_Wolf said: Back after a looong hiatus.
I've got about a dozen cultures sitting in the fridge since 2015. Slants with wooden icecream sticks in them.
It's been eight years. The fridge was fully operational the full time. Any similar experience anyone?
Do you think they are still viable? I'll post pics later today. They are mostly shiitake and various Pleurotus sp.
I found a thread or two with a similar theme, but nobody brought theirs to fruiting from what I've seen.
cheers

Walter_Wolf said: Turns out the dozen cultures I was mentioning were actually 17.

Most of them do look healthy, but my eye in not trained enough to tell for sure. A bigger problem might be the fridge temperature as I measured -1°C in it the next day after the photo session.
How does mycelium handle freezing in anyone's experience?
I will plate them no matter what anyway and report back. My new cleanroom is almost done, so fun times ahead.
cheers everyone

well, this is what they look like four days after plating:

I have growth all over. Even the pioppino which looked awful is growing.
I got me some oats today, so we'll see how far they run. I was not very hopeful because the stuff I transfered was very 'leathery' and didn't look good at all.
what a beautiful day
cheers

Walter_Wolf said: thought one week since plating is a nice benchmark for these vintage cultures. does the growth look promising? ag ag and one of the pulmonarius look weak, but reishi sure is kicking ass.

should the plates be full already? don't think so, right?
will post a week 2 update. will have some grains by then also for sure.
cheers

Walter_Wolf said: So, quick update on the run of the six cultures from 2015, week 3.
We had some cold weather during the week, so the temps were around 18°C. It's now 22. I was observing daily and was kinda worried on the grain colonization speed because there wasn't too much growth visible, but I'm actually pretty pleased now after I checked everything today while taking pictures. I have shaken everything around day 3 or so.
I replated this pulmonarius and this is one week of growth - around 6 or 7 cm. Looks quick enough. I got this one from a buddy as P. pulmonarius 'wawy'. Has anyone heard about this strain ever?
The myc on grain is looking better by the day. I have 14 jars running.

And these are ready to hit the fridge again, poor bastards

Will keep you posted on the progress.
Yeah, the date was actually 11.05. This is 8 days of growth, not 7.
cheers

... and the week 4 update.
I'm a little behind on spawning to bulk, but I got me some straw today, so the Oysters should get onto it during the weekend. All the jars are fully colonized.

I'll be making lime pasteurization because it's the simplest option right now. I have two 20-ish liter plastic drums I'll use and probably just use some trash bags for the rest.
The Reishi I have came with the name 'Lipa clone' so probably using the Lipa's pasteurized fuel pellet tek is the most appropriate for it.
I was supposed to expand some grain to bags (2 x 2,5 kg dry weight), but I burned them in my Presto. Well, live and learn.
Will be updating this thread with the results of this long overdue grow.

Even if I don't have any fruiting bodies to brag about yet, thought a quick update would be nice for now.
So, this is the Reishi culture that was labeled 'Lipa clone' when I got it all this time ago. I thought spawning it into Lipa's pasteurised fuel pellet tek was the most appropriate. This is some 6 days after inoculation:

For the oysters I did the lame lime straw pasteurisation and spawned some two weeks ago into these nice plastic barrels:

Each barrel contains around 3,5 kg dry weight unchopped straw. I had 3 jars of Pl. ost. TSD and Pl. pulm. M33 each containing 120 grams dry weight oats. The HK and Wawy cultures I only had 2 jars each and with just one barrel left, I put them both in. Will be interesting to see what comes out of the holes.
Speaking of which ...

I finally have some visible myc under the micropore tape. Hope all this works out as planned.
In other news, there's no soybean hulls to be found around here. Will have to contact some soybean oil factories I guess because from my understanding they toss all the hulls into the soybean meal byproduct during production.
I got some of the meal stuff and a couple of blocks are ready to get sterilized as we speak. 25% supplementaion rate should work from what I've read.

Awesome to see the slants with Popsicle sticks working so well. I've revived some old cultures (maybe not 8 years old) before but when they're only sitting in agar for years on end its a toss up whether they will rebound or not.

Thanks for the comment, deadmandave. All of the six cultures I plated till now rebounded nicely. Don't know if it has to do with the sticks. They were pretty 'crispy' upon examination. Not sure if the mycelium consumed them or it has to do with them being soaking wet for all this time. Will try to put a piece on agar by itself from the test tubes I still didn't touch, without any visible mycelium on it. We'll see.
The drums I inoculated with the various oysters look pretty much fully colonized:

I don't see any signs of pinning yet, it's been a while since inoculation (a month or so). The outside temp where these have been sitting is around 30°C during the day, so they might be slowed down by the heat. Also the fact that the straw is unchopped could have made the mycelium wanting to 'fill up all the gaps' losing vigor in the process. We'll see what happens in the next week or two.
I do have pins on one of the Phoenixes on a 2,4 (5 lb) block supplemented with 25% soy meal:

Hope these don't abort as my fruiting conditions are far from ideal. Will know in a couple of days.
I also have heavy pinning in the pioppino blocks, made the same way as the phoenix in the previous pic:

I just cut a slit near the tops of the bags to keep the humidity high and allow at least some FAE and evaporation from the blocks. Looks good for now, doesn't it? From my understanding, these don't require huge FAE.
And this is the pasteurized HWFP bag of Reishi:

Taking pics through the bag is less than ideal, but some antlers can be seen through the polypropylene blurr for sure.
This is the current fruiting setup:

I've been waiting for the (single disk) ultrasonic for something like 10 days now. Totally unefficient postal service here.

advise please
would keeping those oyster barrels in the fridge for a day or two help initiate pinning?
thanks
