Clone to Agar Question
    #28376122 - 06/27/23 05:50 AM (1 year, 6 months ago)

06/21 - i cloned a few pins to agar in different plates.

6 days later the pins themselves have fluffed up with myc but still no myc has branched out to the agar surface. Upon observation, a small bot of liquid is visible surrounding the fluffy pins. Almost as if the pins are laying in a tiny puddle of water. Liquid is clear. Is this common when cloning to agar?

A bit on my technique:

-Agar jar was melted back down in microwave
-SAB was cleaned and sanitized. 70% iso sprayed on sides of tub.
-Inside of SAB, agar cooled and plates were poured.
-After Agar reached 80f, fresh pins were harvested and simply dropped onto each plate.
-Within 48 hrs myc was starting to fluff out from pins.

Thats the jist of it.

Able to get pics?
Hopefully just moisture but possibly bacteria, you’ll know eventually but you can always try transferring some of the healthy myc away jic.

Sounds like bacteria either directly from the transfer or from your aseptic technique while transferring it to agar.  You can hope the mycelium makes it past the puddle or take a tiny transfer of the fluffed up growth from the original transfer to a new plate.

Thanks guys, i really suspect that since i didnt do inner tissue cloning and simply dropped the pins on agar thats where it got bacterial and no longer sanitary. Myc growth hasnt done much in the past 4 days or so. Id post pics if necessary but it really sounds like you both got the right idea. Maybe the myc will get past the liquid or if its bacteria maybe it will soon reveal itself. Hoping its just moisture from condensation.
