Questioning Liquid Culture

So I had some jars that myc'd out real fast and ended up forming rafts. I feel like I was only away from them three days without stirring (ofc I forgot stir bars) but I broke up the big chunks best I could with good aggressive "shakes". I had excess LC so it all went in the garage fridge (it runs high, 40ish), in their filtered jars. The fridge isn't immaculately clean (the freezer part anyway) but there's no raw food in it... Anyways, the rafty chunks have sunk to the bottom and we've got some yellowing on the sides. It does just look like mycelial stress, but I'm still pretty amateur, and obvi don't want to 'nocc a batch of garbage... They're one month old, so prob in the fridge 3 weeks. Is it viable y'all think? (These are golden oyster and almond button varieties)

Impossible to say what’s going on in there.  LC should be clear.  .2% nutes is plenty and will allow you to visually inspect them for turbidity (sign of bacterial contamination) and progress.  I’d personally toss those but you can check them on agar if you like.

Don't know what it looked like when fresh or what and how much you've used to make the broth but you made and used LC successfully before? Just asking because you're calling this your excess LC. Doesn't look great to me and additional yellowing that happens after inoculation sounds not good.

My motto with LC is: If in doubt (even the slightest), throw it out.

Ah duh, I do have pics  before it went in the fridge. I understand pics do zero justice to the situation, I figured the consensus would be 'in doubt throw out,' but I had to ask before essentially starting over...

Yes this was extra LC, I have done LC before successfully, but not from jars that have been fridged. I will check on agar, but that affects scheduling and I don't exactly have an HVAC or anything.
I guess I'm mostly wondering: Is there better ways to preserve LC? (Syringing first?) There seems to be conflicting ideas about fridges... and these warm weather strains could probably get away with room temp, I may try that next time. And also, does rafting increase chances of contam? For future reference! Thank y'all

Oof wish I would've found that initially. I have been using other sites for information, since there's such a wealth of information everywhere... Never heard of such low %s. Makes sense, but the reason I have been only doing agar 'on the side' is again, no flow hood. I have had perfect success with LC for months, different species... Guess I've been getting lucky.

Grain to grain without a hood seems foolhardy to me. Am I wrong in this belief?

And working from spores everytime doesn't seem viable for gourmets, schedule wise. Does anybody here do this as their main business?

You can g2g in a SAB but it's not necessary unless you require exponential expansion of a culture. Agar wedges to grain is old reliable. You can grow more than you'll ever consume just with simple a2g.

Agar/plate culture to grain works fine for all species, it even works with bags.

Thanks for phrasing it this way. I definitely at some point fell into the "interested in the destination, not the journey" mindset, when I really do enjoy the whole process. Especially agar work now! SAB makes a nice difference, much better than a fireplace :crazy2:

So what happened with these LCs? Quite simply, some were good and some were bad. Most were good, actually, but there was simply no way to know ahead of time. The one bad culture eventually formed a yellow transparent layer on the surface of the water and would coat the sides of the jar when tilted. It still didn't smell off at all... the nose may know, sometimes but not always. I noticed there was myc growing on the injection port so maybe my new ones will have a tighter seal. And, y'know, remembering stir bars.



But I thought someone out there would appreciate petri pics... I ended up experimenting and just... Transfered the entire mat or mycellium from each variety! I figure if anything would have a better chance to grow it would be pure myc. That, and when I tried to syringe a small amount of LC onto agar, I always fumbled or shat out way too much... Kind of the reason for the delay. Anyway, pics:





These were taken 7.12, not sure what's up with the Citrinopileatus but the tarragon oyster I did ten days prior took over the plate like BLAO! Prompting me to try others. I may just use LC in this manner from here on out, anyone see any problem with this?

I've never seen such badly contaminated broth. Stop using lc.

Yes, I see a problem with growing dense blankets of LC to use as inoculant. This is essentially an agar wedge with extra steps, with an exponentially greater risk for contamination, just use agar. The benefit of liquid culture is it's surface area.

It's possible you've managed, as a new grower who doesn't know anythimg about liquid culture, to have produced a clean batch despite what we're all looking at, but I doubt it.

The proof is in the pudding. Post pics of your spawn.
