Pressure cooking fail
    #28373344 - 06/24/23 08:06 PM (1 year, 6 months ago)

The thing that I was using to elevate the petri dishes vibrated into a hole and fell over and ruined the batch.

I have the same wobbly steamer basket. This was $12 well spent.

It didn't come with a rack other than the one that keeps things a centimeter off the bottom.

is it a rack or a metal type plate thing with holes in it? my presto has the plate type which ya isnt that high up but you can stick metal mason jar lids under that and raise it up so you run your plates.

Stack a couple jar bands or some smashed down aluminum can under the trivet that came with the PC and all will be fine. Use 3 stacks/cans for easy balance.  You could also use 3 half pint jars to space it up. This was a rookie mistake.

Edit: You could probably put the trivet on top of the steamer basket and be fine too.

At very least wrap them in foil. I'd invest in one of these:

https://www.fishersci.ca/shop/products/fisherbrand-sterilizer-box-petri-dishes/03460#

Sterilize them while empty, then pour.

I use five empty cans(from some canned seafood  )  to keep the ideal rack height.

Why not sterilize them after pouring? Then there's no chance of contamination.

Also $130 for some sheet metal is exactly what I expect from science suppliers. Why do they do it?

You can find them cheaper, the link was just for example. Doing "no pour" glass petris is possible but seems like a pita. Pouring plates is easy af, ivenever contaminated a petri dish due to pouring, ever.

Dry sterilization seems the way to go, like pickle rick said.

I'm just thinking that if I have to sterilize the agar anyway, why not do it after pouring.

And you can sterilize in an oven? Why do people use pressure cookers then?

Only the glass petris, you can't use the oven for anything else. Dry heat is ineffective. Dry oven heat is sufficient for the nonporous glass petris, the oven will greatly reduce the risk of condensation.  Sterilizing "no-pour" petris in the pc means you'll have to be very careful not to disturb anything while the agar sets in the pc. You'll likely have very annoying levels of condensation as a result.

I do no pour with PP5 petris. Condensation is really the biggest problem you can run into but I've taken to using a couple of empty plates on the top of each stack for the pc cycle then just leaving the sterilized plates unwrapped in my culture drawer until I use them. They usually dry up in a day or two. Haven't had any contam issues either leaving them unwrapped.

Sterilizing agar is also usually the last thing I do for the day and after I turn the heat off I just go to bed everything is set by the morning. I do wrap the stacks in foil for the pressure cooker cycle just so they are easy to handle. Foil gets carefully removed after and they get gently placed in the drawer.

If you're gonna leave your plates unwrapped don't go waving them around and shit.

No pour makes perfect sense when using pp5 containers. It seems silly with glass petris, but that's my own personal bias, I suppose. They seem too shallow, mostly. The potential for spillage while setting, and massive condensation is high.

Yep, not as clear as glass but overall pretty great. Although someone did show me some very clear pp5 petris recently

They do also get scratched up by scalpels and other tools.
