Help Making syringes!

Im having trouble making sterile syringes. I dont have a Pressure Cooker or glove box and all teks i read are using them. Anyone know a decent method i can use using basic house hold equipment or easy to get shit.
If not any suggestions? like on whole getting spores into PF cake with sterilty?

first take some water and boil it...suck up in the syringe till full..then squirt it out...repeat this 2 more times but the last one keep it in....now wait till this cools(I usually give it overnight just to be sure)..then get your syringe, print, a knife with a pointy tip, tp, a shot glass and a bottle of alcohol...cover the stuff with a t-shirt or a blanket or something....now put on your doctors mask(dry wall mask) and spray the shit outta the room your working in with lysol....give it a few mins to settle then take the alcohol and put some in your hands and move it all around and up your arms...now wipe down the knife real good with alcohol and the needle of the syringe(also go up the syringe just a bit just for good measure) also do the outside of the print if its a tinfoil print...and wipe out the inside and outside of the shot glass...now wait a min or 2 to make sure all the alcohol has evaporated and take your print open it up and scrape with the knife some spores into the shot glass....take the syringe and squirt some of the water into the glass, stir then suck back up...do this a couple times to try and get as many of the spores as possible...and thats it...outta all the syringes I"ve made only 1 has been bad and I think that had to do with how i printed it not the syringe making process

iamthewalrus has it right, except i find that his sterility procedures may be overkill. afoaf just washes his hands and wipes the shotglass/surface of the table he's working over/syringe tip with 99% rubbing alcohol(isopropyl). he also uses the syringe needle to scrape the print into the shotglass instead of the knife!! good luck!

This is a fairly simply method that doesn't use a lot of equipment, assuming you have a shot glass and some isopropyl alcohol (easy to find at any grocery or drug store).

Good point, initially when the oven-tek was first introduced we included it as a portion of our syringe making tek as a way to have a semi-sterile environment at home. Anno was quick to show that it doesn't work and with his evidence we removed it from our tek.
Working in open air with liberal use of a spray disenfectant in a realtively clean environment is usually pretty reliable for most home microscopy.

Ive made a bunch your way with minimal contams....I am really sloppy when making syringes in a fairly dirty environment and have had good sucess

Sure, 71% works well also. There are some reports that it might actually work better but I haven't had the time or the knowledge to explore the science behind those claimes.
Glad to hear our methods are producing good results. We also write our teks with general house-hold usefulness in mind. We all don't have the luxury of nice cleanroom setups and flowhood banks .

I find that using alcohol will dry out the rubber plunger, sometimes making them hard or even crack. This greatly effects the lifespan of the syringe. Not only this, but I also figure a poorly sealing plunger could post a contamination threat. I did use alcohol for a very long time before I figured out what was ruining my plungers. Some of them (actually my large 60ml one I use for small measurements) have become so stiff, it is extremely hard to move the plunger. I would have to say these would be my methods in order of effectiveness:
1. Pressure cooking or autoclaving them. 2. Using a dilute Bleach or 70-90% Alcohol soak (suck and shake). 3. Steaming or boiling 4. Microwaving (extra issues removing and sterilizing the needle separatly) 5. The "suck up boiling water several times" tek.
Pressure cooking allows full sterilization of the entire syringe, and at the same time handling of the syringe is minimized.

70-90%. But I usually get 70%, I never dilute the alcohol. The syringes would last a while, my large 60ml one is so old I don't even remember where I got it. But eventually the plungers become hard, worn, or cracked when I used alcohol. The large one was probably used more than any other for measuring, and most likely has been cleaned with alcohol hundreds of times. This would be my best examples besides some I threw away when I ordered more.
Since I have been simply pressure cooking them the plungers have remained soft and it has to have been close to a year since I ordered them. It really cuts down on the hassle as well since I usually pressure cook water for the syringes as well.

so you use alcohol inside the syringe to sterlize it? I only use alcohol on the needle instead of flame sterlizing...boiling water does the trick for inside
also about my technique being overkill... washing your hands with some alcohol is EASIER than washing them with soap and water and does a better job... and I've used a syringe tip for scraping spores and it don't work nearly as well as the tip of a knife does...you also run the risk of putting a hole in your foil print if your not careful

I just blast the spores off the foil print inside a 1/2 pint jar... No scraping that way

Since you replied to my post I will say NO. I did use alcohol in the past, now I just pressure cook the whole thing. I do not flame the needle, I just wipe with alcohol between each jar.
I use the tip of the needle to gather spores. I can take one swipe and thats all I need. The less the better IMO, provided you get germination.

hey if you get a syringe from a medical source (my mums a nurse)should it already be clean and sterile inside for as long as it remains in the packaging because i can get a hold of syringes easy and and wondered if i would need to go through the process of steilizing or should it just be done any way to make sure and to keep in practice so to speek
