A few basic questions

1) How many transfers is it effective to make with an agar plate, spawn and substrate? (I mean spawn to spawn, substrate to substrate and so on) 2) How is it recommended to store agar plates after inoculation? 3) How to store a full agar plate and how much time can it be stored until the mycelium dies. 4) Is there something special needed to do to make a l.c to agar plate transfer?
I would really appreciate detailed answers

Don't worry too much about making too many transfers. A good practice is to have one dish of which you make a few copies. The master dish you can keep in the fridge wrapped in parafilm and a ziplock. You use one copy to make motherspawn. The motherspawn can inoculate ten times more spawn which is then used to inoculate substrate. Once your copies are finished, you can make new copies from the master dish.
Incubate wrapped in parafilm or clingwrap and in ziplock bag
To store put the package in the fridge
You need a still air box to make clean transfers. Instead of transferring a wedge to another dish, you transfer the wedge to the LC. I stick with a 0.4% malt extract solution. There are plenty other options to be found on the forum.

The problem with LC is you will not know it is bad until you put it on agar or grain and grow it out. If it is bad you have a lot of wasted time there. Time is money and you don't want to waste either. I stick with G2G and just make sure I have enough grain left over after spawning to put the remainder into new grain so I will have it next time I spawn. I use an 8oz spoodle so all my spawn finishes around the same time and I know exactly how much grain I need for a certain amount of bags. Using a spoodle will be more difficult in a still-air box.

I forgot to mention you should make a master slant from your agar plate. It will last longer than plates. Make new masters every year to be safe. To make new masters fill new tubes with agar and transfer from the old master. This is to ensure you have fresh agar. So if you make master slants today make them again on January 1st. Then every year on January 1st make them again it is just easier to do it this way that way you won't forget to do it.
