First attempt at bulk cultivation: mycelium doesn't seem to like millet?
    #28311107 - 05/08/23 07:41 AM (1 year, 8 months ago)

So, despite having had a few issues with my initial spore source, I managed to obtain a cleanly germinated Petri dish. I then proceeded to do one round of transfers out of it, which in turn I used to inoculate 6 grain jars.

I used millet (bought it at a pet store, as bird food) for this first attempt. I let it soak overnight, then drained, put into the jars, and pressure cooked for 2 hours, lids wrapped in foil. Jars have been prepared with a single hole on the lid, filled with polyfill.

Now, after 8 days from the inoculation, the agar pieces are fully colonized and rich in mycelium, but it seems that it doesn't want to move from the agar to the millet, 5 out of 6 jars looks more or less like this (the bottom-right white thing is the light from lamp):

(Yeah, I know, I messed up the amount of grain and they're now too full... still I don't think this is the main issue here.)

While just one looks like this:

They're sitting at room temperature (about 22 Β°C / 72 Β°F). Moisture looks okay-ish too, but I might be wrong. I know that there are plenty of different recipes for the spawn substrate out there, but I would expect to just affect the colonization speed. I hope I have provided enough information, but please let me know if I missed something important about the process.

Is this just a matter of time, or there's something wrong going on here?

You supposed to bury the agar piece deep inside of millet. That's why you need not to fill full jar - shake and bury the agar.

I doubt that's the source of the issue. I leave the agar ontop all the time, myc still colonizes just fine.

@OP, you got pics of the agar you used?

Sure (thanks), so this is the Petri dish I used as a source fro the transfers (the film makes it hard to see unfortunately):

Then pretty much all the transfers looked like this (a couple of days before the inoculation):

The agar is PDA in both cases.

I'm thinking about starting a new batch from another germinated Petri dish that I'm keeping in the fridge for now, but apart form using the proper amount of millet in the jars, I would do pretty much everything else in the same way.

Do you guys think there's something wrong in my process that I need to change?

How big are your holes on your lids?

About 6mm (I guess, 1/4 or 1/5 inches) in diameter.

Can't say anything either way. The wrapped up plate in the first pic seems to have multiple growths on there

I noticed that pattern too, I'm not 100% sure / can't remember, but it could be the result of scratching the agar with the inoculation loop (?).

Assuming the mycelium is OK, what could be the reason for it not jumping straight to the grain? I'm kind of lost here, and not sure of what to try next.

8 days is kind of a long time.. I would give it a shake maybe and see what it does? it cant really hurt at this point..

Tried that as @sonson suggested, but for the sole purpose of trying to bury the agar, or at least to increase the contact surface with the grain, there's otherwise very little to shake.

I decided to try and incubate them at a target temperature of 24 Β°C / 75 Β°F, current room temperature being 22 Β°C / 72 Β°F. Let's see what happens...

For posterity, some of the jars eventually colonized fully, for some reason it was just slow. Higher temperature definitely helped.
